The working principle of graphite digestion analyzer: the sample pretreatment core of "high temperature+strong acid+temperature control" is commonly understood
Graphite digestion analyzer is a core equipment for laboratory sample pretreatment (widely used in food, environment, medicine, metal detection and other fields). Its core function is to decompose solid/semi-solid samples (such as soil, food, ore, biological tissue) into soluble clear solutions through high temperature and strong acid action, which facilitates subsequent elemental analysis using instruments such as atomic absorption spectroscopy and ICP-MS. Its working principle can be summarized as "graphite thermal conductivity+precise temperature control+acid solution digestion+safety protection". The specific disassembly is as follows:
1、 Core foundation: The "efficient thermal conductivity" characteristic of graphite (uniform heat transfer is key)
The core heating component of the graphite digestion instrument is high-density graphite blocks (or graphite tubes, graphite plates), which is the basis of its working principle:
Graphite has the characteristics of high thermal conductivity, strong thermal stability, and uniform heating (superior to ordinary metal heating elements). After being electrified, the graphite block will quickly heat up, and the temperature deviation of the entire heating surface can be controlled within ± 1 ℃ (some models can reach ± 0.5 ℃);
Multiple "sample holes" are reserved on the graphite block for placing digestion tubes (such as polytetrafluoroethylene PTFE tubes, quartz tubes). The sample holes are tightly attached to the digestion tubes, and heat is efficiently transferred through the path of "graphite block → outer wall of digestion tube → sample inside the tube", ensuring that each sample can obtain a uniform heating environment and avoiding local overheating or incomplete digestion.
Simply put, graphite is like a "uniformly heated constant temperature base" that allows all samples to dissolve synchronously under the same temperature conditions, ensuring experimental reproducibility.
2、 Core process: Collaborative digestion reaction of "high temperature+strong acid" (the core of sample decomposition)
The essence of digestion is to accelerate the chemical reaction between strong acid and sample at high temperature, destroy the organic matter and lattice structure in the sample, and dissolve the target elements (such as heavy metals and trace elements) into the acid solution. The specific process is divided into three steps:
Sample and acid mixture: Place the sample to be processed (such as 0.5g soil, 2g food) into a digestion tube and add a suitable acid solution (such as nitric acid, hydrochloric acid, perchloric acid, etc., mixed according to the type of sample). The function of the acid solution is to "dissolve inorganic substances and oxidize organic substances in the sample";
Gradient heating: Insert the digestion tube into the sample hole of the graphite block, and the instrument will gradually heat up according to the preset program (such as heating from 50 ° C to 180 ° C in stages of insulation):
Low temperature stage (50-100 ℃): mainly involves "acid rush" and "pre reaction", removing water and volatile impurities from the acid solution, while allowing the acid solution to come into initial contact with the sample to avoid boiling at high temperatures;
High temperature stage (120-220 ℃, adjusted according to sample type): High temperature accelerates the reaction between acid solution and sample - strong acid oxidizes organic matter (such as proteins and cellulose) in the sample, decomposes the lattice structure of the sample (such as silicates in ores), releases and dissolves target elements in the acid solution, forming a clear digestion solution;
Reaction completion: After a certain period of time (30 minutes to several hours, adjusted according to the complexity of the sample) of high temperature insulation, the solid particles in the sample are completely dissolved, and a uniform and clear solution is obtained in the digestion tube. It can be cooled to a certain volume and used for subsequent detection.
Key logic: High temperature is the "catalyst" that maximizes the dissolution and oxidation ability of strong acids, quickly breaks down the complex structure of the sample, and "releases" the target elements into the solution.
3、 Precise control: intelligent adjustment of temperature and program (ensuring stable digestion effect)
Modern graphite digestion equipment has programmable temperature control function, which is the core to achieve efficient digestion of different samples:
High temperature control accuracy: Real time monitoring of graphite block temperature through built-in temperature sensors (such as platinum resistance PT100), feedback to the controller, automatic adjustment of heating power, ensuring temperature stability at the set value (deviation ≤± 1 ℃), avoiding incomplete digestion or sample loss caused by temperature fluctuations;
Program customization: supports setting multiple heating and holding times (such as "50 ℃ holding for 10 minutes → 100 ℃ holding for 20 minutes → 180 ℃ holding for 60 minutes"), and develops exclusive digestion programs for different samples (such as easily digestible foods and difficult to digest ores) without manual intervention;
Batch processing capability: The number of sample holes on graphite blocks is diverse (commonly 12 holes, 24 holes, 48 holes), and multiple samples can be processed simultaneously. The heating conditions for each sample are completely consistent, ensuring the repeatability and comparability of experimental data.
