Analysis Method of Hyperspectral Fluorescence Testing System
Date: 2025-12-23Read: 2
Hyperspectral fluorescence testing system is an advanced analytical tool widely used in environmental monitoring, biomedical, materials science and other fields. It combines hyperspectral imaging technology and fluorescence spectroscopy technology to provide information on the chemical composition, concentration, and spatial distribution of the sample. The following are some common analysis methods for hyperspectral fluorescence testing systems.
1、 Experimental preparation
Sample preparation
Sample selection: Select appropriate samples based on research objectives, such as plant tissues, cells, chemicals, etc.
Sample processing: In some cases, pre-treatment of the sample is required, such as extracting specific components or preparing thin films, to improve the intensity and clarity of the fluorescence signal.
Equipment Calibration
Light source calibration: Use standard fluorescent materials for calibration to ensure the stability and consistency of the light source.
Spectrometer calibration: Regularly calibrate the spectrometer using a standard spectral source to ensure data accuracy.
2、 Analysis method
hyperspectral imaging
Image acquisition: Obtain the reflection and fluorescence information of the sample at different wavelengths through a hyperspectral camera, forming a hyperspectral data cube.
Data preprocessing: Denoising, background correction, and standardization of the obtained hyperspectral data to improve data quality.
Fluorescence Spectral Analysis
Fluorescence emission spectrum: Record the fluorescence emission spectrum of the sample, analyze its characteristic peaks, fluorescence intensity, etc., and determine the fluorescence characteristics of the sample.
Excitation spectroscopy analysis: By changing the wavelength of excitation light, the fluorescence excitation spectrum of the sample is obtained to further understand the properties of fluorescent substances.
Data analysis methods
Principal Component Analysis (PCA): Using PCA to reduce the dimensionality of hyperspectral data, extract main features, and identify the components of the sample.
Linear Discriminant Analysis (LDA): Used for classification analysis, it distinguishes samples of different categories by constructing discriminant functions.
Support Vector Machine (SVM): Applying machine learning methods such as SVM for sample classification and regression analysis.
quantitative analysis
Calibration curve method: Quantitative analysis is performed by establishing the relationship between the fluorescence intensity and concentration of standard samples.
Multiple linear regression: Use a multiple linear regression model to analyze the concentration of multiple components in a sample.
Spatial distribution analysis
2D/3D imaging analysis: Obtain spatial distribution information of the sample through hyperspectral imaging and analyze fluorescence characteristics in different regions.
Heat map generation: Associate fluorescence intensity with spatial position to generate a heat map for visualizing the fluorescence distribution of the sample.
3、 Interpretation and verification of results
Result visualization
Graphic display: Use chart software to visualize the analysis results, such as drawing fluorescence spectra, heat maps, etc.
Report generation: Organize and analyze data, generate experimental reports, and provide detailed explanations of the experimental process, results, and conclusions.
data validation
Repeated experiment: Conduct multiple experiments to verify the consistency and reliability of the results.
Comparative analysis: Compare with other analytical methods such as mass spectrometry and chromatography to confirm the accuracy of the results.
4、 Precautions
Instrument maintenance: Regular equipment maintenance and calibration are carried out to ensure the accuracy and stability of data.
Light source selection: Choose a suitable excitation light source to avoid fluorescence quenching and other light interference.
Sample stability: Pay attention to the storage conditions of the sample to avoid changes in its properties due to changes in light and temperature.
