Maintenance method of anion exchange chromatography column
1、 Daily operation standards
1. System cleaning and pretreatment
-Cleaning before column connection: Ensure that there are no residual pollutants in the ion chromatograph pipeline and valves, especially removing particles or chemical reagents left over from previous experiments.
-Sample pretreatment: All samples need to be filtered through a 0.22 μ m membrane or centrifuged to remove suspended particles and prevent clogging of the column head sieve plate. For biological samples containing proteins or lipids, it is recommended to use solid-phase extraction (SPE) for additional purification.
2. Pressure and flow rate control
-Strictly follow the maximum pressure tolerance specified in the chromatographic column instructions (usually ≤ 400 bar), as overpressure can cause irreversible damage to the resin.
-Before starting the pump, it is necessary to eliminate bubbles in the pipeline. It is recommended to gradually increase the pressure at a low flow rate (0.3~0.6 mL/min) to avoid bubbles entering the column bed and affecting the separation efficiency.
2、 Regular maintenance and regeneration
1. Rinse and balance program
-Daily rinsing: After daily analysis, rinse the column with deionized water and low concentration alkaline solution (such as 50 mM NaOH) in sequence to remove residual ions.
-Deep regeneration: If the column efficiency decreases, gradient elution method can be used:
-Forward flushing: pure water → acetonitrile (80%) → 0.5 M NaCl solution, each stage lasts for 30 minutes;
-Reverse flushing: Swap the inlet and outlet directions and flush at a flow rate of 0.5 mL/min for 1 hour to remove impurities adsorbed on the sieve plate.
2. Microbial contamination prevention and control
-Rinse the column weekly with a buffer solution containing 0.1% sodium hypochlorite to inhibit bacterial growth. If baseline fluctuations or abnormal pressure increases are found, immediately soak the column core in 70% ethanol solution for 2 hours, and then rinse with sterile water.
3、 Long term storage management
1. Optimization of storage conditions
-Environmental parameters: The storage temperature should be strictly controlled at 15-25 ℃, the relative humidity should be 30% -70%, and direct sunlight should be avoided to prevent resin aging.
-Storage solution selection: It is recommended to use Tris buffer solution (pH 7.0) containing 0.5 M NaCl or 20% ethanol solution to maintain the resin in a moist state and prevent microbial growth.
2. Packaging and labeling
-The two ends of the column need to be fastened with special sealing caps, wrapped in aluminum foil to avoid light, and fixed in the original packaging box.
-The label should indicate the column type, last use date, type of storage solution, and expiration date, and be synchronously entered into the laboratory asset management system.
4、 Special scenario response strategies
1. Accidental drying treatment
-If the column bed is exposed to air for more than 30 minutes, immediately perform the following steps:
-Connect the online degassing device and immerse the column core in pure water at a low flow rate (0.2 mL/min) for 12 hours;
-Gradually transition to high concentration salt solution (1 M KNO3) activation and monitor the recovery of theoretical tray numbers.
2. Strong retention of substances for removal
-For organic pollutants, the "alkali acid" alternating method is adopted:
-First, rinse with 1 M NaOH solution for 2 hours to decompose organic matter;
-Neutralize residual alkaline groups with 0.1 M HNO 3, and finally rinse with deionized water until neutral.
5、 Risk Warning and Life Extension
1. Performance monitoring indicators
-Establish key parameter files: theoretical tray number ≥ 10000 N/m, symmetry factor 0.9~1.5, pressure increase<10% monthly average growth rate.
-When the separation degree drops to 70% of the initial value, start the forced regeneration program.
2. Mechanical damage protection
-It is forbidden to collide, bend or violently vibrate the column. Cushion foam shall be filled and "fragile" shall be marked during transportation.
Through the above systematic maintenance measures, the service life of the anion exchange column can be effectively extended to more than 3 years, while ensuring the accuracy and reproducibility of the separation results.
