Ion exchange chromatography is a separation technique based on the difference in surface charge interactions between target molecules and stationary phases, widely used for the purification of biomolecules such as proteins, nucleic acids, and peptides. However, in practical use, the chromatographic column may experience issues such as abnormal peak shape, retention time drift, and pressure rise, which can affect the separation efficiency.
Typical fault classification and solutions
1. Abnormal peak shape (tailing/forward extension/splitting peak)
-Cause analysis:
-Decreased column efficiency: The aging or contamination of the packing material leads to a decrease in mass transfer efficiency.
-Sample overload: The injection volume exceeds the column capacity, especially sensitive to high valence ions.
-PH/ionic strength mismatch: The mobile phase conditions deviate from the isoelectric point (pI) of the target molecule, leading to an imbalance in adsorption/desorption.
-Solution:
-Optimize sample size: Adjust the sample concentration according to the column volume, usually not exceeding 5% of the column bed volume.
-Adjust the mobile phase: Determine the optimal pH range (within ± 0.5 units) through preliminary experiments, and add competitive ions (such as NaCl gradient elution).
-Cleaning and regeneration: Rinse with low concentration acid/alkali (0.1-1 M NaOH/HCl) or organic solvent (20% ethanol), and reverse rinse if necessary.
2. Retention time drift
-Potential triggers:
-Temperature fluctuations: Changes in environmental temperature affect the diffusion rate of molecules.
-Unstable flow rate: pump pressure fluctuations or bubbles entering the pipeline.
-Collapse of column head: Long term high pressure leads to compression of the filling material.
-Response measures:
-Constant temperature control: Place the chromatography system in a temperature controlled environment (with an accuracy of ± 0.5 ℃).
-Degassing treatment: Ultrasonic degassing of mobile phase, installation of online degassing device.
-Replace the sieve plate: If the column pressure remains high and cannot be restored, it is necessary to disassemble the column and check whether the inlet sieve plate is blocked.
3. Abnormal increase in back pressure
-Mechanical troubleshooting:
-Particle blockage: Improper sample pretreatment introduces particulate matter.
-Microbial growth: Long term storage of aqueous buffer solution can breed bacterial films.
-Disposal process:
-Gradient flushing: Rinse sequentially with deionized water → 5% nitric acid methanol solution → acetonitrile.
-Ultrasonic assisted: Ultrasonic cleaning of metal components to remove attached deposits.
-Replace the protective column: Install a Guard column in front of the analysis column to intercept impurities.
4. Increased baseline noise
-Detector related factors:
-Lamp source attenuation: The decrease in deuterium lamp energy output causes fluctuations in the UV detection baseline.
-Flow pool pollution: Protein residues are adsorbed on the surface of the quartz window.
-Correction method:
-Energy calibration: Run blank control to monitor baseline stability.
-Disassemble and wash the circulation tank: Soak it in dilute nitric acid (pH ≈ 2) and rinse it with ultrapure water.
2、 Deep Maintenance Guide
1. Special scenario handling
-Strong retention of substances: For components with strong hydrophobicity, 6M guanidine hydrochloride+0.1% SDS mixed solution modifier can be used for cleaning.
-Metal chelation effect: Add EDTA (0.5mM) to eliminate metal ion interference.
-Oxidative damage repair: The cation exchange column exposed to air can be reduced to active sites using vitamin C solution.
3、 Preventive measures
1. Sample pre-treatment specifications:
-Centrifugal filtration (0.22 μ m membrane) is used to remove particulate matter.
-Dialysis replacement buffer system to avoid salt precipitation effect.
2. Storage condition management:
-Short term storage: Soak in 20% ethanol solution.
-Long term storage: Dry storage under nitrogen atmosphere.
3. Monitoring of operating parameters:
-Real time recording of column temperature, inlet and outlet pressure difference, and conductivity changes.
-Establish a historical data trend chart to provide early warning of performance degradation.
