What are the aspects to start selecting ACE chromatography columns from
Date: 2025-10-25Read: 0
The selection of ACE chromatography columns should be guided by the separation target, taking into account the characteristics of the stationary phase, physical parameters, and system constraints. It is recommended to prioritize evaluating the properties of the sample and the separation target, then matching the corresponding stationary phase, and verifying the optimization plan through experiments.
The following is an explanation of the selection requirements for ACE chromatography columns:
1、 Clearly define application goals and sample characteristics
Analytical properties
Polarity and solubility: If strong polar compounds (such as peptides and sugars) need to be retained, hydrophilic block stationary phases are preferred; Hydrophobic samples (such as fat soluble vitamins) are suitable for high carbon loading C18-HL columns.
Acidity and stability: Alkaline compounds are prone to secondary interactions with silanol groups, leading to peak tailing. Ultra inert stationary phases (such as C18 series) should be selected, which reduce residual silanol groups and improve separation efficiency through alkaline inactivation treatment.
Separation mode requirement
Reverse phase chromatography: C18 or C8 columns are commonly selected; If the selectivity for aromatic compounds needs to be enhanced, a phenyl column can be used.
Flexibility in method development: For complex mixtures (such as isomers), C18-AR or C18-PFP column fusion offers the advantage of two bonding phases, providing differentiated selectivity.
2、 Selection of core physical parameters
Particle size and column efficiency
≤ 3 μ m particle size: suitable for UHPLC, with the highest column efficiency but significantly increased back pressure.
5 μ m particle size: Balancing column efficiency and system compatibility, it is a commonly used choice for HPLC.
Particle size ≥ 10 μ m: used for rapid screening or preparation grade separation.
Column size design
Length: 150mm is a conventional equilibrium choice, 250mm is suitable for complex samples, and short columns are used for high-speed separation.
Inner diameter: 4.6mm for standard analysis, 2.1mm for microanalysis, and above 10mm for preparation grade.
Aperture adaptability
100 Å: Suitable for small molecules (molecular weight<1000 Da).
300 Å: Large pore size supports the separation of biomolecules such as proteins and peptides.
3、 System compatibility and operational considerations
Equipment adaptability
UHPLC system: sub 2 μ m packing column must be selected to withstand high pressure.
Preparation type HPLC: Choose a large inner diameter column to reduce the risk of blockage.
Mobile phase restriction
High water phase conditions: The C18-AQ column uses polar embedding technology to inhibit hydrophilic interactions and maintain stable retention.
PH value: Avoid using traditional silica gel matrix columns and choose hybrid particle or polymer columns.
