Precautions for operating a spring chromatography column

Spring chromatography columnIt is mainly composed of column tube, piston, spring, screen plate and other components. The principle is to apply fixed pressure to the packing bed through springs, keeping the packing in a tightly packed state, thereby eliminating gaps between the packing and improving column efficiency. This sustained pressure can also prevent column bed collapse and prolong the service life of the chromatographic column.

Spring chromatography columns are widely used in fields such as drug development, disease detection, and biological analysis. They can help researchers screen effective ingredients more quickly, accelerate the development of new drugs, and can also be used for the separation and purification of natural products and active ingredients in biomolecules.

1Pressure and temperature control:

Avoid sudden changes in pressure and temperature, and prevent impulsive or thermal expansion and contraction deformation of the packing inside the column.

When adjusting the flow rate, it should be done slowly, and the rotation speed of the valve during injection should not be too slow.

2Mobile phase and sample processing:

Gradually change the solvent composition to avoid switching directly from organic solvents to pure water (or vice versa).

Filter the sample and flow accordingly to remove insoluble substances and chemical pollutants.

Compounds containing halogen elements (fluorine, chlorine, bromine) may corrode stainless steel pipelines and should be avoided from long-term contact.

3Rinse and store:

Regularly rinse the chromatography column with strong solvents to remove residual impurities.

When storing, the column should be filled with acetonitrile or methanol, and the column joints should be tightened to prevent solvent evaporation and drying.

When not in use for a long time, seal both ends of the column and place it in a dry, dark place.
