How to monitor the effectiveness of high-pressure sterilizers

High pressure sterilizerComposed of sealed barrel body, pressure gauge, safety valve and other components, equipped with electric lock system, bidirectional safety interlock device and intelligent control module, with safety mechanisms such as overpressure protection and low water level alarm. Divided by style into handheld (18-30L), vertical (30-200L), and horizontal types, it supports extended functions such as pulse exhaust and preset programs. During operation, it is necessary to ensure that the sterilized items are properly loaded and the air is exhausted. After sterilization is completed, the lid can only be opened when the pressure returns to zero and the temperature drops to a safe range. Regular maintenance of sealing rings, pressure gauges, and other components is also required.
The monitoring of the sterilization effect of high-pressure sterilizers is a key link to ensure the effectiveness of the sterilization process, mainly divided into three methods: physical monitoring, chemical monitoring, and biological monitoring:
Physical monitoring (process monitoring):
Real time recording of temperature, pressure, and time parameters through the built-in instruments of the sterilizer (pressure gauge, temperature gauge, timing gauge) is required for each sterilization. For example, the lower exhaust sterilizer needs to reach 121 ℃ and 0.1MPa, while the pre vacuum sterilizer needs to reach 134 ℃ and 0.2MPa.
Chemical monitoring:
Indicator card/tape: The chemical indicator card at 121 ℃ or 132 ℃ is placed inside the bag, and the tape is attached outside the bag. The color change verification parameters are up to standard (such as 20 minutes at 121 ℃).
B-D test: specifically designed for pre vacuum sterilizers to detect residual cold air, which needs to be conducted daily before sterilization.
Biological monitoring:
Use thermophilic Bacillus subtilis (ATCC 7953) as an indicator bacterium at least once a week.
Operation steps:
Place the biological indicator at 5 points in the sterilization cabinet (upper, middle, lower layers).
After sterilization, culture for 48 hours. If the spores survive (the culture medium turns yellow), sterilization fails.
