Shodex OHpak LB-800The series is a water-soluble size exclusion chromatography column. Suitable for the separation of various water-soluble polymers, proteins, oligomers, and the determination of molecular weight distribution of polymers. By minimizing column loss as much as possible, the baseline noise when used in conjunction with a light scattering detector is effectively reduced, resulting in more accurate determination of molecular weight.LB-800We have a range of products with different exclusion limits, which can be selected based on the molecular weight range of the samples.

Conditions for using chromatographic columns
Non ionic hydrophilic samples and ionic hydrophilic samples
Salt solutions and buffer solutions are often used as mobile phases. Typical representatives include the following:
salt solution----Sodium chloride aqueous solution, sodium nitrate aqueous solutionNa2SO4水溶液、硫酸钙水溶液、硫酸铵水溶液
buffer solution----The concentration of salt added to phosphate buffer, hydrochloric acid buffer, acetic acid buffer, and citric acid buffer is0.05~0.3 M.
(Attention)
(1)pHPlease control the value range within3.0~10.0.
(2)If there are chloride ions in the mobile phasepHPlease be higher than6.
(3)The maximum salt concentration of the solvent that can be used is0.5 M.
(4)It is not recommended to use boric acid buffer because the functional groups in the filler will react with boric acid.
Non ionic hydrophobic samples and ionic hydrophobic samples
To suppress the hydrophobic adsorption of hydrophobic samples, it is recommended to add organic solvents to the mobile phase. In addition, ionic samples also require the addition of salt solutions.
Organic solvents that can be added, please refer to the instruction manual for details.
(Attention)
When replacing the mobile phase in the chromatographic column with a mobile phase containing organic solvents, please control the flow rate at0.5 mL/minfollowing.
UseDMSOWhen, the flow rate must be controlled within0.3 mL/minThe temperature of the chromatographic column must be higher than50 ℃.
Protein samples
Urea and6MGuanidine aqueous solution can be used as a protein denaturing agent and as a mobile phase. Samples with low solubility in water, such as membrane proteins, are added to a solventSDSorBrij -35Waiting for surfactants, the effect is very good.
(Attention)
Because a high concentration of denaturing agent is required, frequent solvent changes in the chromatographic column may shorten its lifespan. Therefore, it is recommended to use specialized chromatography columns.
