By pointing the mouse cursor to a trajectory, you can see the precise parameter values of the selected sperm, and in a pop-up window, you can see the classification results. The video can be replayed, backtracked, and played every frame. It can be saved without compression (in full quality) or compressed using software on the computer. You can also adjust the detection threshold or change the classification criteria.

The number of sperm detected and tracked will be displayed on the screen, and it can be checked whether enough cells have been tracked (WHO reference is 200). When the analysis is completed, the results of automatic classification and manual counting are automatically transmitted to the database.

Sperm head morphology is an important criterion for accurate diagnosis. The software is set up for analyzing static images, and the samples need to be stained with Diff Quik dye according to strict Krueger standards. We chose Diff Quik, which is a rapid staining method for sperm. Through Diff Quik, the sperm head is stained light blue in the acrosome region and dark blue in the posterior acrosome region, which is a good foundation for precise graphic analysis. The following parameters were measured for each sperm:

The software automatically classifies sperm into normal and head pathology based on head parameters. You can easily manually correct the results and develop other abnormalities (tail pathology, neck pathology) if you cannot obtain good staining images for automatic detection. For example, due to incorrect sample preparation, there is a special tool that can manually animate cell contours.

Nowadays, it is common to record the number of morphological sperm defects divided by the number of defective sperm, which is called the teratozoospermia index (TZI). After the analysis is completed, TZI is automatically calculated. The TZI value should be between 1.00 (each malformed sperm has only one defect) and 3.00 (each malformed sperm has head, neck, and tail defects).

The analysis results can be printed out in HTML format. The report is generated based on the database and saved in the corresponding database records. Sports analysis requires special counting plates with fixed depths such as Leja, Makler, MicroCell, Cell Vu, etc. These counting boards provide standard volumes and sperm are distributed within a single layer of aggregation plane.

Practice has shown that Leja counting slides are the preferred option for CASA.

Leja provides a wide range of disposable counting plates for sperm analysis. Slips with 2 or 4 chambers, with chamber depths of 10 or 20 micrometers. Leja slides have a standard low abundance quantitative series that can detect low concentrations of sperm. Its diagnostic significance is that you can choose from IUI, IVF, or ICSI to determine the threshold. Leja slides are very easy to use, and new members of the laboratory can quickly master how to use them.

Description:

Makler sperm counting plate (chamber) is a convenient device used for rapid and accurate sperm counting, motility, and morphological evaluation, using undiluted samples.

It consists of two parts (Figure 1):

1. The main part below consists of a metal substrate (A) and two handles (H). There is an optical flat glass flat disk (D) at the center of the substrate, on which the sample is placed. There are four needles (P) around the flat plate. Their * is 10 microns higher than the flat plate.

2. The upper part is covered with glass (C) surrounded by a metal ring. There is a 1 mm2 grid in the center of its lower surface, divided into 100 grids, each measuring 0.1 X 0.1mm. When the cover glass is placed on four needles, the space enclosed by 10 squares on a row is one millionth of a milliliter. Therefore, the number of sperm heads in the 10 squares represents their concentration in millions/mL.

You can choose between bright field or phase difference. The software can recognize dark targets on a bright background or vice versa. So dark fields can also be used.

The 20X objective lens is used for mobility. If a reusable counting board, such as Makler, is used, a long working distance lens (approximately 1.8 millimeters) is required. If a disposable counting board, such as Leja, is used, it is not necessary.

For sperm morphology, analyzing static color images with a resolution of 1280X1024 directly in computer memory also improves performance.

The camera can record the settings applied in motion and morphology, and save them in its own memory. So if you replace the computer, there is no need to reset it

Sample preparation

Sperm concentration is measured using a special counting plate, which has a fixed depth to provide a standard volume, and the sperm are distributed in the same focal plane and can move freely. The tracked sperm is always in the focal plane. The sperm counting plate can use undiluted samples. Be careful when mixing sperm samples during preparation to avoid foaming. If the sample is highly viscous, it can be aspirated several times with a syringe to make it uniform.

In order to obtain high-quality sperm staining images for morphological evaluation, semen slurry should be diluted first and removed after centrifugation. The sperm balls are resuspended within the volume to achieve the highest sperm concentration, but not exceeding 80 x 10 ^ 6/ml. 0.2 to 0.5ml of semen, depending on semen concentration, is diluted to 10ml with physiological saline at room temperature. The test tube is centrifuged at 800g for 10 minutes and the majority of the suspension settles. The ball is suspended in the remaining saline solution, usually 20-40ul, and gently tilted through a test tube. Then hang 5-10 ul of this

(a) Feather method dilutes semen. The droplets spread along the back edge of the tilted glass slide and are pushed forward to the plane of the slide to form a layer.

(b) Wash the sample using the pipette method. A droplet of suspended matter diffuses onto the surface of a glass slide through a horizontal pipette.

Prepare the coating. Make the least two coatings from fresh semen samples to repeat the experiment. Firstly, the bottom of the slide should be cleaned, washed with 70% ethanol and dried before placing small drops (5 to 20ul) of semen. If the semen concentration is greater than 20X10 ^ 6/ml, 5ul can be used; if the semen concentration is lower than 20X10 ^ 6/ml, 10 to 20ul should be used. The feather method uses the edge of another slide to spread the droplets along the surface, but care must be taken not to make the coating too thick. Feather technique works well when viscosity is low, but it is usually not suitable for viscous semen. As an alternative, a drop of semen can be prevented from being in the middle of the slide, and then another slide can be placed upside down to spread the semen between them: then gently pull apart both slides to make two coatings at the same time.
