Validation of intracellular RNA protein binding: RIP
Experimental significance
Studying the binding of RNA and protein in cells is helpful to understand the dynamic process of the post transcriptional regulatory network, discover the regulatory targets of miRNA, understand the overall level of RNA changes and changes in the post transcriptional regulatory network such as malignant tumors and genetic diseases, and apply them to pathological diagnosis and scientific research.
Experimental principle and steps
The basic principle of RNA Binding Protein Immunoprecipitation (RIP) is to use antibodies against the target protein to capture endogenous RNA protein complexes, then separate the RNA protein complexes together through immunoprecipitation, and finally use RT-PCR RIP Seq (high-throughput)
Sequencing or RIP Chip (microarray) are used to identify the bound RNA sequence. The RIP process is shown in the following figure
