Trypsin is a serine protease that specifically cleaves the carboxyl terminus of lysine and arginine residues. The strict specificity of trypsin is the basis for protein identification. Natural trypsin is prone to self hydrolysis, producing pseudotrypsin, which has a wide specificity and contains chymotrypsin like activity. These self hydrolysis products present in trypsin preparations can generate excess peptide fragments, thereby interfering with the data analysis of fragments detected by mass spectrometry. Sequencing Grade Trypsin can provide maximum specificity. The lysine residues in pig trypsin have undergone reductive methylation modification, becoming highly active and stable molecules with particularly strong resistance to self hydrolysis. The specificity of purified trypsin is also affected by TPCK treatment, which leads to the loss of chymotrypsin

The processed trypsin is purified by affinity chromatography and made into freeze-dried powder, which can tolerate mild denaturation conditions such as 0.1% SDS, 1M urea, etc. It can also maintain 50% activity in 2M guanidine hydrochloride. Product Description:

*Trypsin can specifically hydrolyze the carboxyl terminal peptide bonds of lysine and * residues. Untreated * will self hydrolyze, forming fragments that interfere with protein sequencing, HPLC, or mass spectrometry analysis of peptide chains. In addition, self hydrolysis will lead to the production of false *, exhibiting additional * characteristics. Promega has undergone methylation reduction reaction modification, which to some extent prevents self hydrolysis. In the functional stability test, the activity retained by modified * after incubation at 37 ℃ for 3 hours is at least twice that of unmodified *.
The modified sequencing grade * was further processed with TPCK and purified by affinity chromatography to obtain highly active and stable molecules. Sequencing grade modification * is provided in the form of 20mg freeze-dried powder per bottle, which is convenient to use and comes with a stability optimized resuspension buffer. Suggest using protease: protein ratio of 1:100 to 1:20 (w/w).
