Leica paraffin sectioning machines (such as Leica RM2235, RM2255, RM2265 series, etc.) are key equipment used for tissue sectioning in pathology laboratories. Their maintenance directly affects the quality of sectioning, equipment lifespan, and experimental efficiency. The following are detailed maintenance methods, covering daily cleaning, key component maintenance, regular calibration, and safety precautions:
1、 Daily cleaning and basic maintenance
1. Clean immediately after slicing
Knife holder and blade:
After each slice, use a soft bristled brush or air balloon to remove paraffin debris from the knife holder to avoid residue affecting the accuracy of the next slice.
If disposable blades are used, they should be replaced in a timely manner; When reusing the blade, gently wipe the blade with a cotton ball dipped in 75% ethanol (do not touch the blade directly with your hands).
Sample head and fixture:
Clean the paraffin residue on the sample head (such as tissue clamp) to prevent uneven slice thickness caused by unstable tissue fixation.
Wipe the surface of the sample head with an anti-static cloth to prevent static electricity from adsorbing dust.
Workbench:
Clean up the scattered paraffin debris during the slicing process to prevent it from entering the mechanical transmission parts (such as guide rails and gears).
2. Daily maintenance before shutdown
Turn off the power and gas supply:
Cut off the power supply of the slicer, and if equipped with a cooling system (such as a cooling table), turn off the cooling function.
Drain the compressed air pipeline (if any) to avoid corrosion of components by condensed water inside the pipeline.
Dust protection:
Cover with a dust cover to reduce dust from entering optical components (such as microscope interfaces) or mechanical gaps.
2、 Deep maintenance of key components
1. Knife holder and injection system
Tool holder lubrication:
Add 1-2 drops of specialized lubricating oil (such as Leica recommended silicon-based lubricating oil) to the sliding guide rail of the tool holder every month, and move the tool holder several times to evenly distribute the oil film.
Avoid using regular engine oil to prevent the adhesion of paraffin or the attraction of dust.
Sample wheel cleaning:
Remove the sample wheel (rotating part of the sample head), wipe the wheel shaft and gear with a soft cloth, and remove the accumulation of paraffin.
Check the elasticity of the injection wheel spring. If it is loose, it needs to be replaced to ensure stable tissue clamping force.
2. Mechanical transmission system
Guide rails and sliders:
Wipe the X/Y axis guide rails quarterly with dust-free paper dipped in isopropanol to remove oxide layers and dirt.
Apply a thin layer of lubricating grease (such as Molykote 33) again to reduce friction resistance.
Handwheel and gearbox:
Check if the handwheel rotates smoothly. If there is any jamming, disassemble the gearbox and clean the internal gears (operated by a professional engineer).
The oil level of the gearbox should be kept in the middle of the scale line, and the gear oil should be replaced every 2 years (such as Shell Omala S2 G 150).
3. Cooling system (if equipped with a cooling platform)
Cold table cleaning:
Wipe the surface of the cold table with a soft cloth every week to avoid difficulty in removing paraffin after freezing.
Check if the condensate drain pipe is unobstructed to prevent water from flowing back to the circuit board.
Refrigeration efficiency monitoring:
Record the temperature of the cooling table every month (usually set to -15 ℃ to -20 ℃). If the temperature rises too quickly, check the refrigerant pressure or whether the condenser fins are blocked.
3、 Regular calibration and performance verification of Leica paraffin slicer
1. Slice thickness calibration
Standard tools:
Use thickness calibration plates (such as 5 μ m, 10 μ m, 20 μ m standard plates) to verify the deviation between the actual thickness of the slice and the set value.
Calibration steps:
Install calibration plates onto the sample head and set the target thickness (e.g. 10 μ m).
10 consecutive slices, measure the thickness of each slice with a micrometer, and calculate the average value.
If the deviation is greater than 1 μ m, adjust the thickness parameters through slicing machine software or mechanical fine-tuning knob.
2. Horizontal and Vertical Inspection
Levelness:
Place the level on the worktable of the slicer, adjust the bottom support foot to center the bubble, and ensure that the slicing plane is perpendicular to the blade.
verticality
Use a ruler to check the perpendicularity between the tool holder and the workbench, with a deviation of less than 0.5 °. Otherwise, adjust the fixing screws of the tool holder.
3. Electrical safety testing
Grounding resistance:
Every year, use a grounding resistance tester to test the grounding wire of the equipment, and the resistance value should be less than 4 Ω to prevent the risk of leakage.
Insulation performance:
Measure the insulation resistance between the power cord and the equipment casing with a megohmmeter, which should be greater than 1M Ω to ensure safe operation.
4、 Long term disuse and storage
1. Preparation before discontinuation
Cleaning:
Disassemble all detachable components (such as tool holder, sample head) and use an ultrasonic cleaning machine (if available) to remove paraffin residue.
Wipe the mechanical transmission parts and apply rust proof oil (such as WD-40).
Moisture proof treatment:
Place desiccants (such as silicone bags) inside the device, close all openings to prevent moisture from entering and causing circuit corrosion.
2. Storage environment requirements
Temperature and humidity:
Store in a dry (relative humidity<60%), ventilated room, away from direct sunlight or high temperatures (>30 ℃).
Regular power on:
Every 3 months, power on and run for 1 hour to drive mechanical components and prevent lubricating oil from solidifying.
5、 Safety precautions
Operational safety:
When replacing the blade, it is necessary to wear anti cutting gloves and use a specialized blade gripper to avoid direct contact with the blade by hand.
During the slicing process, it is forbidden to insert hands or tools into the movement area of the knife holder to prevent injury.
Chemical safety:
The solvents such as ethanol and isopropanol used for cleaning should be kept away from sources of fire and operated in a fume hood.
Waste disposal:
Discarded blades should be placed in a dedicated sharps box and disposed of according to medical waste regulations to prevent the risk of stabbing.
6、 Professional maintenance suggestions
Annual major overhaul:
Contact authorized Leica engineers for comprehensive disassembly and cleaning, replacing aging components such as O-rings and belts.
Software upgrade:
Regularly update slicing machine control software (such as Leica CARE Suite), fix vulnerabilities, and optimize slicing parameter algorithms.
Through systematic maintenance, Leica paraffin slicer can maintain high-precision slicing performance for a long time (thickness error<0.5 μ m), reduce failure rates (such as knife jamming and thickness drift), and extend equipment service life to more than 10 years. It is recommended that the laboratory develop a maintenance schedule and record each maintenance data for traceability and analysis.
