Isolation of vascular endothelial cells: Isolation and culture of rat cardiomyocytes are crucial for studying cardiomyocyte contractility, myocardial hypertrophy, and heart failure. Rat cardiomyocytes are firmly connected by intercalated discs, making isolation of rat cardiomyocytes highly challenging compared to neonatal mouse cardiomyocytes. We have developed a self built perfusion system that can effectively isolate adult rat cardiomyocytes. After treatment with calcium ions, the isolated rat cardiomyocytes can be used in a wide range of experiments, including drug screening.

Isolation of vascular endothelial cells

Vascular endothelial cells are arranged on the inner surface of the entire cardiovascular system as a single layer of flat epithelium. They serve as the boundary cells between the vascular wall and blood, and form the morphological basis for the formation of a closed cardiovascular duct system. At present, it is believed that endothelial cells in the blood vessel wall have secretion and synthesis functions, which are related to the coagulation system, fibrinolysis system, and complement system. The isolation and culture of endothelial cells in large blood vessels (such as umbilical vein, large artery, etc.) mainly use digestive enzyme perfusion method, while the isolation and culture of endothelial cells in microvessels (such as skin, brain, spinal cord, etc.) are relatively difficult and have their own characteristics. Endothelial cells are important tools for studying vascular function, tissue repair, regenerative medicine, aging, and tumor metastasis in vitro.
