| I. Background | |
| The characteristic of cancer-related inflammation is the presence of specific inflammatory cells and mediators, including cytokines and chemokines. Recent evidence suggests that the signal transduction and transcription activator (STAT) family proteins, particularly STAT3, play a crucial role in selectively inducing and maintaining the pre cancerous inflammatory microenvironment during the onset of malignant transformation and cancer progression. Signal transduction and transcription activator 3 (STAT3) is a transcription activator that directly transmits signals from extracellular receptors to the nucleus, mediates the expression of some genes, and plays an important role in cellular processes such as apoptosis. Under the action of cytokines and growth factors, STAT3 is phosphorylated by receptor associated Janus kinase (JAK), forming homodimers or heterodimers, and translocated into the nucleus to act as a transcriptional activator. STAT3 signaling is the main intrinsic pathway of malignant tumors, as it is frequently activated in malignant cells and plays a critical role in regulating many genes crucial for cancer inflammation in the tumor microenvironment. | |
