STING1(stimulator of interferon genes 1)是先天免疫信号通路的核心调控因子,属于G蛋白偶联受体超家族成员,主要通过cGAS-STING1信号轴感知细胞质异常DNA(如病毒、细菌或线粒体DNA),结合cGAMP后激活TBK1-IRF3及NF-κB通路,诱导I型干扰素(IFN-I)和促炎因子(如TNF-α、IL-6)的产生,从而介导抗病毒、抗肿瘤免疫应答及炎症调控。其功能异常与多种疾病密切相关:在自身免疫疾病中,STING1突变(如N188H)或UXT缺失可导致I型干扰素过度激活,引发系统性红斑狼疮(SLE)、皮肤血管病变(SAVI)等;在肿瘤中,STING1表达水平与预后呈双向关联,此外,STING1突变或表达水平亦可作为SLE等疾病的诊断标志物及肿瘤预后评估指标[1-2]。
