Overview

This reagent is made using the principle of colloidal gold immunochromatography assay (GICA). After the sample is added to the loading well, it moves along the chromatographic membrane with the colloidal gold label. If H9 avian L antibody is present in the sample, it binds to the colloidal gold label and the antigen on the detection line, displaying a purple red color. If H9 avian L antibody is not present in the sample, no color reaction occurs.

【 Principle 】

This reagent is prepared using the principle of colloidal gold immunochromatography assay (GICA). 【 Storage and Validity Period 】Store in a cool and dry place (2-30 ℃), with a shelf life of 24 months.


1. This test card is only applicable for testing poultry serum/whole blood.

2. Serum: Extract 2-3ml of blood according to conventional methods and place it in a clean and dry test tube. Let it stand for about 1 hour until the blood coagulates, then centrifuge at 4000 rpm for 10 minutes (the blood can also be left to stand for about 2 hours until it coagulates and naturally forms serum), and separate the serum. Require clear serum without hemolysis or contamination. Serum samples can be stored at 2-8 ℃ in the short term, but need to be stored at -20 ℃ in the long term.

3. Whole blood: Fresh anticoagulated whole blood should be collected and used on the same day, while whole blood without anticoagulants should be collected and used immediately.


1. Serum/whole blood sampling method: Tear open the aluminum foil packaging bag of the detection card, take out the detection card, place it on a flat and clean table, use a pipette in the bag to slowly drip 1 drop of blood serum or 2 drops of whole blood into the sampling hole, and then drip 2 drops of diluent into the sampling hole.

2. Place at room temperature for 10-15 minutes to determine the result, and the color development result after 15 minutes is invalid.

1. Negative: In the observation hole, only a purple red line appears in the control line area (C).

2. Positive: In the observation hole, both the detection line area (T) and the control line area (C) show purple red lines. The higher the antibody titer, the darker the color of the detection line (T).

3. Invalid: In the observation hole, there is no purple red line in the control line area (C).

The detection threshold of this reagent is 1:32 for HI test, so by diluting the tested serum by multiple ratios and finding the positive * dilution ratio, the antibody titer can be determined. For example, the serum to be tested is diluted with physiological saline at ratios of 1:2, 1:4, and 1:8, respectively. Then, three samples with different dilution ratios are tested. If both 1:2 and 1:4 are positive and 1:8 is negative, then the positive * dilution ratio is 1:4. The final antibody titer of this sample is 25 (1:32) multiplied by 22 (1:4), which is 27 (1:128).


Explanation of Experimental Results

For immunized samples, the level of antibodies reflects the effectiveness of the immune response. For samples that have not been immunized, a positive result indicates the possibility of H9 subtype avian L virus infection, which needs to be analyzed in conjunction with clinical and other methods.


Limitations of Experimental Methods

This test card is for screening purposes only and should not be used as a basis for diagnosis.


【 Precautions 】

1. Please read the instructions carefully before the experiment. The various reagents provided by this product are only for use in this experiment.

2. Do not use non sample required liquids such as animal serum as negative controls.

3. Expired or damaged aluminum foil bags are not allowed to be used.

4. When removing the detection card from the refrigerator, it should be restored to room temperature before opening. The opened detection card should be used as soon as possible to prevent it from becoming ineffective due to moisture.

5. Avoid using samples that are contaminated, turbid, severely hemolytic, or have a large amount of blood lipids.

6. Avoid contact with the chromatography membrane at the sample well and detection window on the detection card.

7. The waste from the experiment should be treated as pollutants and properly disposed of in accordance with local regulations.
