【 Principle of Inspection 】
Fluorescein can bind to beta polysaccharides in fungal cell walls, such as cellulose and chitin, but cannot bind to tissue cells. Fluorescein induces fluorescence in various filamentous fungi and yeasts under ultraviolet excitation, including Candida, Bryum, Sphingomonas, Microsporomycota, Histoplasma, and Aspergillus. Potassium hydroxide in the dye solution can dissolve keratin in the sample tissue, eliminate impurities, and promote the addition of solvents to dissolve keratin in the sample tissue more quickly, making the tissue more transparent and improving detection sensitivity. The selected background counterstaining solution can reduce the background brightness of the sample tissue, making the fluorescence signal clearer.

【 Main Ingredients 】
The composition of fungal fluorescent dyes from different brands may vary slightly, but they are usually mainly composed of fluorescent dyes and solubilizers. Some products also contain KOH, which can be used to dissolve keratin and help observe fungal structures more clearly.

1. Dandruff, nail shavings, and hair specimens: First disinfect the affected area with 75% alcohol, and then use a blunt surgical knife to scrape off an appropriate amount of dander or nail shavings; Use tweezers to extract several diseased hairs and place them on a glass slide.
2. Preparation of tissue smear or pathological section involves steps such as fixation, dehydration, and dewaxing prior to routine pathological staining.

1. Place the specimen on a clean glass slide;
2. Drip the dye solution to cover the entire specimen;
3. Cover the glass slide and wait for 1 minute;
4. Remove excess dye and observe the glass slide under a fluorescence microscope.

Clinical Application
Dermatology: Used to detect fungal infections such as tinea pedis, tinea corporis, and tinea pedis.
Respiratory department: assists in diagnosing pulmonary fungal infections by detecting fungi in sputum.
Obstetrics and Gynecology: Used for detecting gynecological secretions to help diagnose various vaginal infections.
Other departments: In ophthalmology, otolaryngology and other departments, it can be used to detect fungal infections in the eyes, external auditory canal and other areas.
