Overview

The PCR product amplified by high fidelity DNA polymerases with 3 '→ 5' exonuclease activity such as Pfu, HiFi, Vent, or KOD is a flat end DNA fragment without an A tail. Operation steps:

1. Flat end PCR products are purified by PCR cleaning (non-specific amplification) or gel recovery (non-specific amplification).

2. The purified flat end DNA fragment is added with A tail according to the following reaction system:

3. Gently mix and keep at 72 ℃ for 20-30 minutes.
