Design, layout, and construction of high-throughput gene sequencing laboratories in medical institutions
High throughput sequencing testing laboratory, also known as NGS laboratory, uses high-throughput sequencing technology as a detection method to complete clinical disease screening, diagnosis or auxiliary diagnosis, tumor gene mutation detection and other projects.
Due to the extensive amplification of nucleic acid templates detected by NGS technology, laboratory contamination can easily lead to inaccurate test results; Moreover, NGS experiments have high requirements and multiple influencing factors, and improper handling of the experimental process can also lead to abnormal detection results. Therefore, a strict, orderly, and standardized NGS laboratory is essential.

Design requirements for high-throughput sequencing laboratory
According to the requirements of the "Work Standards for Clinical Gene Amplification Testing Laboratories" issued by the National Health and Family Planning Commission, in principle, clinical gene amplification testing laboratories should set up the following areas: reagent storage and preparation area, specimen preparation area, amplification area, and amplification product analysis area. These four areas must be physically independent of each other, and each area should always be in a separated state, whether in space or in use, without direct air communication. According to the functionality of the instrument used, the areas can be appropriately merged. For example, using a real-time fluorescence PCR instrument, the amplification zone and amplification product analysis zone can be merged; By using an automated analyzer that integrates sample processing, nucleic acid extraction, and amplification detection, the specimen preparation area, amplification area, and amplification product analysis area can be merged.
The airflow in the clinical gene amplification laboratory can be arranged according to the reagent storage and preparation area, specimen preparation area, amplification area, and amplification product analysis area to prevent amplification products from entering the pre amplification area along the airflow. It can be carried out in a manner where the air pressure decreases from the direction of the reagent storage and preparation area, specimen preparation area, amplification area, amplification product analysis area. This can be achieved by installing exhaust fans, negative pressure exhaust devices, or other feasible methods. If conditions permit, it is recommended to control the working temperature of the sequencing room where the NextSeq CN500 sequencer is located at 18-22 ℃ and the humidity below 65%.
Strong and weak electrical requirements for high-throughput sequencing laboratories
weak current:
Suggest reserving 1 network port and 1 telephone port for each room. And equip the high-throughput sequencing laboratory with an exclusive network of over 8M.
Strong electricity:
The distribution of electrical appliances is roughly as follows:
Reagent preparation room: refrigerator, freezer, etc; Total power ≤ 5kw
Sample preparation room: high-speed centrifuge, low-speed centrifuge, vertical mixer, etc; Total power ≤ 10kw
Amplification room: PCR instrument, fluorescence quantitative PCR instrument, etc; Total power ≤ 5kw
Sequencing room: UPS power supply (both sequencer and server are connected to UPS power supply); UPS connection power 6KW
