After sterile treatment, the reagent does not contain nitrogen sources such as ACC. DF medium is often used in combination with ADF medium to analyze the ACC deaminase characteristics of bacteria. The growth of the strain in ADF medium is better than that in DF medium, indicating that the strain can grow on ACC as a nitrogen source, that is, it can produce ACC deaminase.

Self provided materials:

1. ADF medium, NDF medium (optional)

2. Aseptic centrifuge tube or culture vessel

3. Vaccination Ring

4. Shake the bed

5. Colorimetric cup

6. Spectrophotometer

DF culture medium manufacturerSteps for operating the culture medium (for reference only):

1. Take sterile centrifuge tubes or culture vessels and add 3ml of DF medium, ADF medium (optional), and NDF medium (optional).

2. Inoculate the purified strain simultaneously into the DF medium, ADF medium (optional), and NDF medium (optional) mentioned above, and shake on a shaker at 150r/min for 72 hours.

3. Observe the growth of the same strain in three different media: DF medium, ADF medium (optional), and NDF medium (optional).

4. Measure the OD values of each culture medium at 600nm using a spectrophotometer to assess the growth of the bacterial strains.
