The discovery of transcription factor binding sites (TFBS) has always been a research hotspot in functional genomics and epigenetics studies. The traditional ChIP seq (chromatin immunoprecipitation sequencing) method can effectively detect TFBS with good antibody quality. However, good antibodies are hard to come by, which limits the wider application of ChIP seq.
The emergence of DAP seq technology has made TFBS research no longer limited to species or restricted by antibody quality, providing a new and effective tool for the study of transcription factors in the field of life sciences.
DAP seq has the same functionality as ChIP seq. By using in vitro protein expression technology, labeled transcription factors are expressed and bound to genomic DNA libraries in vitro. Then, all DNA binding to transcription factors is isolated, and high-throughput sequencing is used to identify the binding sites of transcription factors.
