Macroporous adsorbent resin, also known as fully porous resin or polymer adsorbent, is a type of polymer adsorbent that does not contain ion exchange groups and has a macroporous network structure. It belongs to porous cross-linked polymers and has the function of separating and enriching organic matter. It is generally white, milky white, or yellow particles, and some new resins are yellow, brownish yellow to reddish brown in color, with particle sizes typically ranging from 20 to 60 mesh. Physically and chemically stable, insoluble in water, acids, bases, and hydrophilic organic solvents, insoluble when heated, and can be used below 150 ℃. The use of macroporous adsorption resins to separate and enrich alkaloids from natural products is often based on the adsorption capacity and elution rate of alkaloids, and is selected from many common types of macroporous adsorption resins. In the experiment of separating and purifying total alkaloids from Zanthoxylum bungeanum, Lu Shihui et al. gradually reduced the ethanol volume fraction and loaded it onto a macroporous resin column for ethanol volume fraction gradient adsorption, overcoming the defects of sedimentation, insoluble sedimentation, and easy column blockage that occur after the ethanol extract is recovered in one go. While enriching total alkaloids, they also achieved the goal of preliminarily separating each component according to their polarity, which is the same as gradient elution. In general, the alkaloid extracts obtained by commonly used methods currently contain not only alkaloids and their salts, but also many other lipid soluble components or water-soluble impurities that require further purification treatment. Compared with other purification methods, other purification methods have low power, complex operation, and high cost. The eluent used in the macroporous resin method is ethanol of different concentrations, which has lower toxicity and is easy to operate. Therefore, the macroporous resin method is an ideal method for separating and extracting alkaloids.

Saponins are widely present in natural products and are a class of chemical components with various pharmacological effects such as lowering blood sugar, regulating blood lipids, antiviral, tumor suppression, and immune regulation. By using static adsorption and desorption methods, and optimizing temperature and ethanol concentration, D101 improved the purification rate of saponins from Polygonatum sibiricum from 6.7% to 32.35% in 7 different macroporous resins, with a recovery rate of 85.47%, providing a method for isolating saponins from Polygonatum sibiricum and other plants. In the study of total saponins, it was found that impurities, especially fat soluble impurities such as chlorophyll, contained in D101 resin during separation and purification can contaminate and block the resin. Therefore, pre-treatment of the drug solution is necessary to remove these impurities. Cheng et al. purified total saponins from Tribulus terrestris using macroporous resin, achieving recovery and purification rates of 87% and 68%, respectively. In summary, by carefully selecting the resin type, setting the process parameters reasonably, and using macroporous resin to refine saponins, good selectivity, high saponin yield, and high purity can be achieved.
