The type I mouse tail collagen provided by Qishi Biotechnology was prepared by Birkedal Hansen's method using steps such as acetic acid extraction and sodium chloride precipitation. Our company's mouse tail collagen can be used to coat cell culture vessels and cultivate cells that are difficult to adhere to in ordinary cell culture vessels. It can also be used for the preparation of three-dimensional gels, simulating the growth of cells in a real three-dimensional environment. This product has been validated through long-term testing and can stably and efficiently promote cell adhesion growth.

Instructions for Use

Recommended dosage for general packages:

Dilute the original solution before use, and the dilution ratio should be determined according to the experimental requirements. The surface coating of cell culture vessels is generally recommended to have a concentration of 1-5ug/cm2Dilute the original solution with 20mm acetic acid solution to the required multiple before coating, let it sit for 1-2 hours, and then aspirate the excess liquid.

Taking a coating concentration of 2 μ g/cm2 as an example, first dilute the original solution with 20mm acetic acid solution to 12ug/ml, and then add it to the corresponding culture vessel according to the following volume:

Preparation of three-dimensional collagen:

When the concentration of type I mouse tail collagen is greater than 1mg/ml and the pH is around 7.0, a three-dimensional gel with certain strength can be formed. It is recommended that the gel concentration be 1-2mg/mL. This product is dissolved in a 20mm acetic acid solution and needs to be neutralized by adding 0.2 x volume of 0.1mol/L NaOH during the gelation process.

Required solution (sterile and pre cooled): 10 x PBS (containing 10 mg/L phenol red for pH indication) or 10 x culture medium, 0.1 mol/L NaOH, double distilled water

A.Preparation of three-dimensional collagen without cells(To prepare1mL1mg/mLTaking 3D glue as an example)

1) Add 200 μ L of LI type mouse tail collagen (5mg/mL) to a centrifuge tube placed in an ice bath, and add 660 μ L of H2O。

2) Then add it to 40 μ L of 0.1mol/L NaOH (if 12 μ L of 40mol/L NaOH is added to the collagen solution in reverse, local collagen coagulation may occur due to the inability of NaOH to mix quickly), mix immediately.

3) Add 100 μ L of 10 × PBS or 10 × culture medium, mix well, and immediately add to the culture vessel (pH around 7 after mixing).

4) Place the culture vessel at room temperature (around 25 degrees) for 20 minutes until the gel solidifies, then transfer it to the culture chamber. If 10 × PBS is used in the preparation, an appropriate volume of cell culture medium needs to be added for pre equilibration before use.


B.Preparation of three-dimensional collagen containing cells(To prepare1 mL1mg/mLTaking 3D glue as an example)

1) Prepare the cells suspended in the culture medium and place them in an ice bath.

2) Add 200 μ L of LI type mouse tail collagen (5mg/mL) to 40 μ L of 0.1mol/L NaOH (if 40 μ L of 0.1mol/L NaOH is added to the collagen solution in reverse, local collagen coagulation may occur due to the inability of NaOH to mix quickly), mix immediately.

3) Add 23 μ L of 10 × PBS or 10 × culture medium and mix well (pH around 7 after mixing).

4) Add 737 μ L of cell suspension, mix well, and immediately add to the culture vessel.

5) Place the culture vessel at room temperature for 20 minutes until the gel solidifies, then add an appropriate volume of cell culture medium and transfer it to the incubator for cultivation.

After receiving the product, store it in the dark at 2-8 ℃ with a shelf life of two years and cannot be frozen;
