Calcein, chemical formula C30H26N2O13, is a bright yellow powdery organic fluorescent compound with CAS number 1461-15-0. Its sodium salt forms orange red crystals and is easily soluble in water, emitting green fluorescence [1] [3]. Its melting point exceeds 300 ℃, and it needs to be sealed and stored at room temperature to maintain stability. This substance is synthesized by reacting fluorescein with reagents such as iminodiacetic acid and formaldehyde. It is mainly used as a chelating indicator and fluorescent indicator for titration analysis and fluorescence detection of metal ions such as calcium, strontium, barium, copper, and manganese. It is also used in the biomedical field for live cell staining and drug delivery system evaluation [2] [5].

In the 1970s, calcein was introduced into soil exchangeable calcium determination, overcoming the limitation of traditional indicators being adsorbed by magnesium precipitation.
