The principle of Nessler's reagent colorimetric method:Under alkaline conditions, ionized ammonium in water is converted into free ammonia, and then a certain amount of Nessler's reagent is added. Free ammonia reacts with Nessler's reagent to form a yellow complex. The analytical instrument measures the absorbance A of the reaction solution at a wavelength of 420nm, and the standard working curve is queried based on the A value to calculate the ammonia nitrogen content.
The principle of salicylic acid colorimetric method:In the presence of sodium nitroprusside, free ammonia and ammonium ions in the sample react with salicylate and hypochlorite ions to form a blue compoundMeasure the absorbance A at 670nm, query the standard working curve from A, and calculate the content of ammonia nitrogen. The salicylic acid colorimetric method has advantages such as sensitivity and stability. Adding potassium tartrate masking can remove the interference of cations, especially calcium and magnesium ions.
