Overview

ADF medium is mainly composed of phosphate, glucose, gluconic acid, citric acid, etc., and contains numerous trace elements such as manganese, copper, iron, zinc and other metal ions. After sterile treatment, the reagent contains ACC (also known as 1-aminocarbonyl-1-cyclopropanecarboxylic acid).

Self provided materials:
1. DF medium, NDF medium (optional)
2. Shake the bed
3. Colorimetric cup
4. Spectrophotometer
5. Aseptic centrifuge tube or culture vessel
6. Vaccination Ring
Operation steps (for reference only):
1. Take a sterile centrifuge tube or culture vessel and add 3mlADF medium、 DF medium (optional), NDF medium (optional).
2. Inoculate the purified strains simultaneously onto the above mentioned strainsADF medium、 Place in DF medium (optional) and NDF medium (optional) and shake on a shaker at 150r/min for 72 hours.
3. Observe the growth of the same strain in three different media: ADF medium, DF medium (optional), and NDF medium (optional).
4. Measure the OD values of each culture medium at 600nm using a spectrophotometer to assess the growth of the bacterial strains.
Result:
When the strain is placedADF mediumWhen the growth of the strain is significantly better than that of DF medium, it indicates that the strain can grow on ACC as a nitrogen source, that is, the strain can produce ACC deaminase.
Notes:
1. Cultivating in DF medium or NDF medium is an optional step, not a mandatory step.
2. For your safety and health, please wear lab coats and disposable gloves when operating. Pay attention to aseptic operation and avoid microbial contamination.
3. If there is no spectrophotometer, a regular enzyme-linked immunosorbent assay (ELISA) reader can also be used for measurement.
