Human pluripotent stem cells (hPSCs) require additional extracellular matrix to adhere and grow in a non feeder culture system. The initial experiments of hPSC cell culture were conducted on Matrigel, a substrate preparation extracted from EHS tumors in mice. Matrigel hPSC maintenance medium can effectively support the long-term proliferation of hESC, but it is a complex protein mixture from mice that can easily cause variations in the culture system. Due to the variability and heterogeneity of matrix adhesives, their use in regenerative medicine applications is complicated. Research has found that the combination of collagen IV, fibronectin (FN), laminin (LN), and vitronectin (VTN) supports stable long-term proliferation of hESCs in stem cell culture. As alternative methods, laminin (LN), glass fiber and fibronectin, as well as synthetic polymers, have been identified as directions for future substitution of Matrigel

Laminin 521 is a key cell adhesion protein in the natural stem cell niche. Recombinant human Laminin-521 is a non animal based recombinant heterotrimer composed of three chains: alpha 5, beta 2, and gamma 1. Laminin521 can bind to various receptors on the cell surface, including integrins, polysaccharides, etc. Among them, integrin α 6 β 1 binds to Laminin with high affinity and is the main cell adhesion receptor of Laminin, directly determining Laminin's support for cell adhesion ability. Integrin α 6 β 1 is highly expressed in hESC and hiPSC. LN521 binds to cell surface receptors, activating cellular signaling pathways and producing more functional cells.
