What are the specific methods for using a pipette correctly and how should one pay attention to them? Pipettes are precision instruments and equipment, and attention should be paid to their accuracy and precision. They should also be maintained and serviced regularly. 1. Adjust the range
When adjusting the range, if you want to switch from a large volume to a small volume, follow the normal adjustment method and rotate the knob counterclockwise; But if you want to adjust from a small volume to a large volume, you can first rotate the scale knob clockwise to the scale beyond the range, and then adjust it back to the set volume, which can ensure the maximum height measured. During this process, do not turn the button out of range, otherwise it may get stuck in the internal mechanical device and damage the pipette.
2. Assemble the gun head (suction nozzle)
The correct method to lift the pipette tip onto the liquid gun is to insert the pipette vertically into the tip and slightly rotate it left and right with a little force to ensure a tight fit. If it is a multi-channel (such as 8 or 12 channels) pipette, you can align the * channel of the pipette with the * nozzle, then insert it at an angle, and shake it forward or backward to tighten it. The sign of the gun head clamping is slightly beyond the O-ring, and a clear sealing ring can be seen formed at the connecting part.
Three methods of pipetting
Before pipetting, ensure that the pipette, pipette tip, and liquid are at the same temperature. When sucking liquid, keep the pipette upright and insert the tip of the gun 2-3 millimeters below the liquid level. Before suctioning, you can first suck and release the liquid several times to wet the suction nozzle (especially when you need to suck viscous or liquid with a density different from water). At this point, two methods of pipetting can be employed.
One is the forward pipetting method. Press the button with your thumb until the * stop point, then slowly release the button to return to the original position. Then press the button until the * stop point to discharge the liquid, pause for a moment and continue pressing the button until the second stop point to blow out the remaining liquid. The second method is reverse pipetting. This method is generally used for transferring high viscosity liquids, bioactive liquids, easily foaming liquids, or extremely small amounts of liquids. The principle is to first inhale more liquid than the set range, and there is no need to blow out residual liquid when transferring the liquid. First press the button until the second stop point, then slowly release the button until it reaches the origin. Then press the button to the * stop point to discharge the liquid with the set range, continue to hold the button at the * stop point (do not press it down again), remove the gun head with residual liquid, and discard it.
4. Correct placement of pipettes
After use, it can be hung vertically on the pipette rack, but be careful not to fall off. When there is liquid in the pipette tip, do not place the pipette horizontally or upside down to avoid liquid backflow and corrosion of the piston spring.
