Two types of pipetting methods that can be used with a pipette
A pipette is a device used for quantitative transfer of liquids, which has the advantages of flexible use, easy maintenance, compact size, and convenient portability. It is widely used in fields such as biology, chemistry, and scientific research. The pipette requires daily maintenance during use to ensure the normal operation of the instrument.
Basic points of pipette:
1. Note that the pipette, suction nozzle, and liquid are at the same temperature.
2. Pay attention to tightly fitting the suction nozzle onto the connector.
3. When suctioning, the pipette should be kept upright with the suction nozzle only a few millimeters below the liquid level.
4. Pre wash the suction nozzle before suction, that is, suck and release the liquid 5 times. This is particularly important when transferring liquids that are viscous or have a density different from water.
5. Always use your thumb to control button movement to ensure consistency and coherence.
Before pipetting, ensure that the pipette, pipette tip, and liquid are at the same temperature. When sucking liquid, keep the pipette upright and insert the tip of the gun 2-3 millimeters below the liquid level. Before suctioning, you can first suck and release the liquid several times to wet the suction nozzle (especially when you need to suck viscous or liquid with a density different from water). At this point, two methods of pipetting can be employed.
One is the forward pipetting method. Press the button with your thumb until the * stop point, then slowly release the button to return to the original position. Then press the button until the * stop point to discharge the liquid, pause for a moment and continue pressing the button until the second stop point to blow out the remaining liquid. The second method is reverse pipetting. This method is generally used for transferring high viscosity liquids, bioactive liquids, easily foaming liquids, or extremely small amounts of liquids. The principle is to first inhale more liquid than the set range, and there is no need to blow out residual liquid when transferring the liquid. First press the button until the second stop point, then slowly release the button until it reaches the origin. Then press the button to the * stop point to discharge the liquid with the set range, continue to hold the button at the * stop point (do not press it down again), remove the gun head with residual liquid, and discard it.
Correct placement of pipette: After use, it can be hung vertically on the pipette rack, but be careful not to fall off. When there is liquid in the pipette tip, do not place the pipette horizontally or upside down to avoid liquid backflow and corrosion of the piston spring.
