In the Good Manufacturing Practice (GMP) system, controlling the moisture content of raw materials, intermediates, and finished products is of crucial importance. Moisture not only affects the stability, effectiveness, and safety of drugs, but may also pose risks such as microbial growth, chemical degradation, or changes in physical properties. Therefore, accurate, reliable, and traceable moisture determination methods are one of the key steps in GMP compliance. Among numerous moisture determination techniques, the Coulomb method moisture analyzer is widely used in quality control laboratories in the pharmaceutical industry due to its high sensitivity, high accuracy, and good reproducibility, and has become an important tool for moisture analysis in GMP environments.
1、 Principle of Coulombic Moisture Determination
The Coulombic method for moisture determination is based on the Karl Fischer (KF) reaction principle, which quantitatively determines the moisture content in the sample through electrochemical means. Unlike the volumetric method, the Coulomb method does not rely on volumetric titration of titrants, but instead generates iodine through electrolysis of iodide. Iodine reacts with water in a fixed stoichiometric ratio, and the moisture content is calculated based on the amount of electricity consumed during the electrolysis process. Due to the fact that 1 mol of water corresponds to 2 Faraday charges (i.e. 192970 coulombs), the instrument can accurately convert the mass of water in the sample based on Faraday's law. This method is particularly suitable for the determination of trace moisture (usually with a detection limit of up to 1 μ g H ₂ O), and is very suitable for quality control of moisture sensitive APIs and excipients.
2、 GMP requirements for moisture determination
GMP regulations (such as Chinese GMP, EU GMP, FDA cGMP) have clear requirements for analytical methods, including method validation, system suitability testing, data integrity, instrument calibration and maintenance, etc. Specifically for moisture determination, the following key points need to be met:
1. Applicability of the method: The selected method must be applicable to a specific sample type and accurately reflect its true moisture content;
2. Accuracy and precision: The results should have good repeatability and reproducibility;
3. Data reliability: All raw data, audit trails, and electronic records must comply with the ALCOA+principles (attributable attributes, clarity, synchronicity, originality, accuracy, completeness, consistency, and persistence);
4. Equipment validation and calibration: The moisture meter needs to undergo installation confirmation (IQ), operation confirmation (OQ), and performance confirmation (PQ), and be calibrated regularly;
5. Preventing cross contamination: The sample processing process should avoid introducing external moisture or residual interference.

5. Fast and efficient: A single measurement is usually completed within a few minutes, increasing laboratory throughput.
4、 Usage precautions and compliance recommendations
Despite the obvious advantages of Coulomb method, the following points still need to be noted in GMP environment:
Solvent selection: Suitable KF solvents (such as methanol, ethanol, or specialized composite solvents) should be selected based on the solubility of the sample to avoid side reactions;
Sample processing: Solid samples should be thoroughly ground to ensure complete moisture release, while liquid samples should avoid moisture absorption;
System sealing: The instrument must maintain good sealing to prevent interference from environmental moisture;
Regular maintenance: electrode cleaning, electrolyte replacement, drying tube replacement, etc. need to be included in the preventive maintenance plan;
Personnel training: Operators need to receive dual training on GMP and instrument operation to ensure consistency in method execution.
