Calibration cycle and accuracy verification method for online sampling analysis system

  Online sampling analysisAs a key component of Process Analysis Technology (PAT), the real-time data of the system directly affects the determination of reaction endpoints, quality control, and continuous manufacturing decisions. To ensure the reliability of data, it is necessary to establish standardized calibration cycles and accuracy verification mechanisms.
1、 Recommended calibration cycle
Key processes (such as sterile formulations, high activity API synthesis): Conduct system suitability testing before each batch and perform comprehensive calibration every 3-6 months;
Conventional R&D or pilot testing scenarios: It is recommended to calibrate every 6-12 months. If the system stability is good and the historical data deviation is less than 2%, it can be appropriately extended;
Sensor components (such as pH and conductivity probes): need to be calibrated with standard buffer solution before each use;
Chromatographic/spectroscopic detectors (such as online HPLC, FTIR): Perform daily performance checks (DPC) and annual metrological calibration as recommended by the instrument manufacturer.
2、 Accuracy verification method
Standard substance comparison method: Inject a known concentration of standard solution into the system, compare the online analysis results with the theoretical value, and calculate the recovery rate (which should reach 98-102%);
Repeatability test: Take the same sample 5 times in a row, and the RSD should be ≤ 2%;
Delay time measurement: Measure the time lag from sampling to peak output through step response experiments to ensure timely process control;
Multi point linear validation: Verify the linear correlation coefficient (R ² ≥ 0.999) at low, medium, and high concentrations.
3、 Data integrity and compliance requirements
All calibration and validation data must be stored electronically and comply with the requirements of 21 CFR Part 11 or EU GMP Annex 11;
Calibration certificates should be issued by institutions with CNAS qualifications and traceable to national standards;
The system software needs to have an Audit Trail function to record all parameter modifications and operation logs.
