【 Instrument Encyclopedia 】 Principle of Drug Hot Melt Extruder and Analysis of Its Application Advantages in the Pharmaceutical Field
Date: 2025-11-10Read: 3
  Drug hot melt extruderIt is an efficient equipment that continuously prepares uniform drug formulations by extruding and mixing thermoplastic materials at high temperatures through screw extrusion. In recent years, with the development of continuous pharmaceutical technology, the application of hot melt extrusion technology in advanced drug formulations such as solid dispersions, sustained-release formulations, nanomedicines, and combination drugs has become increasingly widespread, especially in improving the solubility and bioavailability of insoluble drugs and achieving precise drug delivery.
1、 Basic principle of drug hot melt extruder
Hot Melt Extrusion (HME) is a continuous processing technology based on thermal and mechanical energy. Its core is to heat materials such as drugs and polymer carriers to a molten or softened state under solvent-free conditions. Through the shearing and mixing action of screws, they are evenly dispersed or dissolved. Then, they are extruded through molds and solidified by cooling to obtain the desired drug formulation.
1. Main workflow
1. Feeding
Feed the active pharmaceutical ingredients (APIs) with polymer carriers (such as PVP, PEG, Eudragit, etc.), plasticizers, stabilizers, and other excipients into the extruder hopper through gravity feeding or forced feeding systems.
2. Heating&Melting
The material enters a barrel with a temperature control jacket and is heated to a set temperature (usually 100-200 ° C, depending on the material properties) through electric heating or hot oil circulation;
Under high temperature, thermoplastic polymers and some drugs melt or soften, forming a melt with certain fluidity.
3. Screw shearing and mixing
The molten material is subjected to strong shear, tensile, and mixing forces under the rotating push of a multi-stage screw (with conveying, compression, mixing, and dispersing functions);
The different configurations of screw components (such as conveying screw blocks, mixing screw blocks, kneading blocks, etc.) can achieve uniform dispersion of materials, molecular level mixing, molecular dispersion or microcrystalline dispersion of drugs in carriers;
For poorly soluble drugs, this step can promote their formation into amorphous or molecular states dispersed in the polymer, significantly improving their solubility.
4. Extrusion&Forming
The uniformly mixed melt is extruded through a die to form continuous strip-shaped, thin film, rod-shaped, or granular intermediates;
The extruded material is cooled (air-cooled/water-cooled), pelletized or further processed into final dosage forms such as tablets, capsule fillers, implants, orally disintegrating tablets, sustained-release scaffolds, etc.
2. Key technologies and parameter control

parameter
effect and influence
temperature control
The melting state of materials, drug stability, and compatibility are affected, and precise temperature control is required in each area
screw speed
Balancing mixing efficiency and thermal degradation risk is necessary to affect shear strength, mixing effect, and residence time
Feeding speed
Control the residence time of materials in the barrel, affecting mixing uniformity and production capacity
Screw design
Different functional segments (conveying, mixing, dispersing) determine the mixing effect and material form control
pressure control
Affects extrusion stability and mold forming effect
2、 Analysis of the application advantages of drug hot melt extruder in the pharmaceutical field
Hot melt extrusion technology has significant application advantages in modern pharmaceutical industry due to its solvent-free, continuous, efficient mixing, and controllable drug release characteristics, especially in solving difficult problems such as insoluble drugs, slow-release and controlled release systems, and compound formulations.
1. Improve the solubility and bioavailability of poorly soluble drugs (key applications)
✅ Challenge: Most new drugs are BCS Class II (low solubility, high permeability) or Class IV (double low) drugs, with poor oral bioavailability;
✅ HME Solution:
By hot melt extrusion, poorly soluble drugs can be highly dispersed in hydrophilic or enteric polymer carriers in molecular or amorphous form (such as PVP, HPMC, Soluplus, Eudragit E, etc.);
The formed solid dispersion can significantly increase surface area, inhibit drug crystallization, and accelerate dissolution rate;
• Effectively improve drug solubility and oral absorption, and enhance bioavailability (some drugs can increase several times or even tens of times).
🔬 Application examples: Solubilizing agents for BCS Class II drugs such as antifungal drugs, anticancer drugs, antiviral drugs, NSAIDs, etc.
2. Preparation of sustained-release drug formulations
✅ HME can achieve uniform mixing of drugs and polymers, accurately controlling drug release behavior:
By selecting polymers with different solubility, permeability, or degradation rates (such as Eudragit RL/RS, PLGA, polyvinyl alcohol, etc.), the release rate of drugs in the gastrointestinal tract can be controlled;
During the extrusion process, the drug and carrier form a homogeneous matrix, which achieves controlled release effects such as zero order/first order/pulse release through diffusion, dissolution, or permeation mechanisms;
Suitable for special drug release systems such as long-acting implants, transdermal patches, gastric retention systems, and colon targeting.
🔬 Application examples: long-term psychotropic drugs, pain management sustained-release agents, diabetes drug controlled release system.
3. Achieve continuous, efficient, and scalable production
✅ Compared with traditional batch wet granulation, solvent evaporation, melting spray and other processes:
HME is a continuous production process with no solvents or drying steps, resulting in a shorter process and higher efficiency;
Easy to scale up production (from laboratory to pilot to industrial production), with stable process parameters and good repeatability;
Reduce batch differences, improve product quality consistency, and be more suitable for cGMP and QbD (quality comes from design) concepts.
🔧 Advantages: Suitable for intelligent, continuous, and flexible production needs in the context of pharmaceutical industry 4.0.
4. Suitable for heat stable drugs and excipients, expanding the space for drug formula design
HME does not require the use of organic solvents, avoiding solvent residue, toxicity, environmental pollution, and recycling issues;
Suitable for drug and polymer systems with good thermal stability, by reasonably controlling temperature and residence time, various new drug delivery formulations can be expanded;
Flexible introduction of plasticizers, stabilizers, surfactants, pigments, taste masking agents, etc. to optimize formulation performance.
5. Preparation of special dosage forms and complex drug systems
Oral solid dispersions, sustained-release pills, implants, oral disintegrating tablets, multi-layer composite tablets, 3D printed drug materials;
Preparation of nano drug carrier precursors, uniform blending of compound drugs, and basic materials for targeted delivery systems;
Provide technical support for personalized medicine, precision medicine, and new drug delivery methods.
