What are the advantages of a fully automated PCR machine?
Date: 2025-05-18Read: 0
A fully automatic PCR instrument is an experimental device used for DNA amplification, which achieves rapid replication of DNA sequences by heating and cooling at different temperatures to assist the action of specific enzymes. The emergence and development of PCR instruments have greatly improved the speed and accuracy of DNA analysis, and are widely used in fields such as molecular biology, medicine, forensics, and environmental science.

  Fully automatic PCR machineWorking principle:
1. Denaturation process
In the first step of PCR, the template DNA is heated to high temperature (usually 94-98 ° C) to break down the double stranded DNA into two single strands. This process needs to be completed in a short period of time to ensure complete separation of the DNA strands.
2. Annealing process
When the temperature drops to 50-65 ° C, the designed primers will bind to single stranded DNA, forming short double stranded DNA structures. This step is called the annealing process, which provides a starting point for the extension of the next amplification stage.
3. Extension process
In the extension step, the temperature is usually adjusted to 72 ° C (suitable for the working temperature of common Taq enzymes), and by adding DNA polymerase and appropriate nucleotides, the synthesis of DNA chains starts from the primer starting position, amplifying more DNA molecules.
4. Loop process
These steps involve multiple cycles, typically 20-40 times, with the ultimate goal of amplifying the target DNA fragment to a sufficient amount for subsequent analysis. It can automatically control these temperature cycles to ensure accuracy and repeatability of each step.
Advantage:
1. High degree of automation
It can automatically complete the entire process from sample heating, temperature control, sample amplification, data collection, etc., greatly reducing manual intervention and improving the efficiency and accuracy of experiments.
2. High throughput
Supporting high-throughput operations and being able to process a large number of samples simultaneously has significant advantages for large-scale clinical testing, disease screening, and other applications. The high-throughput PCR instrument can detect 96 or even more samples at once through a microplate.
3. Easy to operate
Users can input experimental parameters through a computer or touch screen, and once set, the instrument will automatically run and complete various tasks. No manual intervention is required, thus reducing the possibility of operational errors.
4. Improve accuracy and repeatability
Being able to accurately control temperature changes and reaction time ensures consistency in experimental conditions, improving the accuracy and repeatability of experiments.
5. Save time
In traditional PCR methods, operators need to manually change the temperature, monitor the experimental process, and usually require a longer reaction time. Can accelerate these processes and improve overall experimental efficiency.
Application areas of fully automatic PCR machine:
1. Medical diagnosis
PCR instruments are widely used in the medical field, especially in genetic testing, pathogen detection, and tumor gene mutation screening.
2. Forensic identification
In forensic identification, PCR technology is commonly used in fields such as paternity testing and autopsy. By extracting DNA samples from the deceased or suspect and amplifying them using a PCR machine, DNA information can be quickly obtained for case investigation.
3. Environmental monitoring
In environmental monitoring, PCR technology can be used to detect harmful substances or pathogenic microorganisms in water and air. Being able to quickly detect a large number of environmental samples helps in timely warning and handling of pollution incidents.
4. Agriculture and Food Safety
In the field of agriculture, PCR technology can be used for rapid detection of pathogenic microorganisms during crop cultivation. In the food industry, PCR can be used to detect the presence of pathogenic microorganisms or illegal additives in food to ensure food safety.
5. Scientific research applications
Widely used in scientific research fields such as genomics, transcriptomics, and molecular biology. It is of great significance in gene expression research, genomics analysis, and disease mechanism research.
