Optimization of cleaning strategy for organic solvents and cell culture residues in fully automatic bottle washing machine
Date: 2025-09-23Read: 2

The optimization of cleaning strategies for organic solvents and cell culture residues in fully automatic bottle washing machines can be achieved from three aspects: chemical cleaning agent adaptation, physical cleaning parameter regulation, and programmed intelligent control. Combined with the actual needs of the laboratory, efficient, safe, and traceable cleaning effects can be achieved.

1、 Optimization of cleaning strategy for residual organic solvents
Selection of chemical cleaning agents
Residual organic solvents (such as xylene and ethanol) require the use of specialized cleaning agents with low foaming and strong emulsifying ability. For example, a solution containing disodium ethylenediaminetetraacetic acid (EDTA Na ₂) can effectively decompose lipid soluble stains after heating and boiling; Alkaline cleaning agents (such as 5% -10% trisodium phosphate solution) remove oil residues through saponification reaction. The fully automatic bottle washing machine needs to be equipped with an automatic liquid addition system to accurately control the concentration of cleaning agents according to the cleaning program, avoiding manual addition errors.
Physical cleaning parameter regulation
Residual organic solvents require a high-temperature and high-pressure environment to accelerate dissolution. The fully automatic bottle washing machine can set a cleaning temperature of 70-95 ℃, coupled with a 0-1000L/min circulation pump to drive a 360 ° rotating spray, forming a high-pressure water flow of over 70bar, and deeply peeling off residues in the dead corners of the bottle dish. For example, for the slender structure of the micro pipette tip, the spraying time needs to be extended to more than 5 minutes to ensure the solvent is discharged after emulsification.
2、 Optimization of cleaning strategy for residual cell culture
Selection of chemical cleaning agents
Cell culture residues (such as proteins and cell debris) require enzymatic cleaning agents. For example, urea detergent can dissolve protein structures, while specialized cleaning agents containing proteases can efficiently decompose organic matter at 40-60 ℃. The fully automatic bottle washing machine needs to support multiple cleaning programs. First, soak the residue in enzyme cleaning agent at low temperature to soften it, then switch to alkaline cleaning agent to neutralize acidic substances, and finally rinse with pure water.
Physical cleaning parameter regulation
Precision vessels such as cell culture bottles should avoid mechanical damage. The fully automatic bottle washing machine should adopt flexible spray technology, which reduces the impact force on the bottle wall by adjusting the spray pressure to 30-50 bar. At the same time, increase the number of rinses to at least 3 times and use RO water or WFI water (injection water) to reduce ion residues, ensuring compliance with sterile requirements for cell culture.
3、 Programmed intelligent control optimization
Preset cleaning program library
The fully automatic bottle washing machine needs to be equipped with specialized programs such as "organic solvent cleaning" and "cell culture bottle cleaning", covering the needs of fields such as biology, chemistry, and medicine. For example, for residual organic solvents, the program can set a four stage process of "pre washing (room temperature) - alkaline washing (80 ℃) - rinsing (90 ℃) - drying (70 ℃)"; For cell culture bottles, the "enzymatic hydrolysis (40 ℃) - alkaline washing (60 ℃) - pure water rinsing (3 times) - sterile drying" mode is used.
Data traceability and verification
Real time detection of rinse water quality through conductivity monitoring system to ensure residual ion concentration<1 μ S/cm. At the same time, the equipment needs to be equipped with a printer component to automatically record cleaning time, temperature, concentration and other parameters, generate reports that comply with GMP/GLP standards, and meet audit traceability requirements.
