Principles and Application Guidelines for Drug Melting Point Determination Technology

The determination of drug melting point is one of the crucial physical analysis methods in drug quality control, purity identification, and process research. The results are directly related to the safety, efficacy, and stability of the drug.
1、 Technical principles
The essence of melting point is the temperature at which a substance transitions from a solid state to a liquid state. For pure matter, this transformation occurs at a very sharp temperature point. The core technical principles are based on the following two points:
Purity indication principle: According to the laws of physics and chemistry, pure crystalline substances have a fixed and sensitive melting point. If the drug contains impurities, it will cause its melting point to decrease and the melting distance (the temperature range from the beginning of melting to melting) to widen. Therefore, by observing changes in melting point, the purity of the drug can be quickly evaluated.
Crystal identification principle: The same drug molecule may form different crystal structures (i.e. "polymorphs"). Different crystal forms have different melting points and physicochemical properties, which directly affect the solubility, bioavailability, and stability of drugs. Melting point determination is a commonly used method for preliminary screening and identification of different crystal forms.
At present, capillary method is a standard method specified in pharmacopoeias of various countries, such as the Chinese Pharmacopoeia, USP, and EP. The operation is to fill a fine dry sample into a thin-walled capillary tube with one end closed, attach it to a thermometer, place it in a heating bath (such as silicone oil), and heat it at a constant speed at a specified rate. The initial melting and full melting temperatures are determined by visually observing phenomena such as sample collapse and droplet formation.
Modern instruments commonly use the hot stage method, which places the sample on a controllable hot stage with a temperature sensor, observes the melting process through a microscope, or uses a photoelectric sensor to automatically detect changes in transmittance to determine the melting point, with higher automation and accuracy.
2、 Application Guide
Melting point determination is widely used throughout the entire lifecycle of drug development and production
Raw material quality control: As a key physicochemical constant, the melting point is included in the quality standards of raw materials. The measurement results need to be consistent with the melting point of the known pure product to ensure inter batch consistency and purity meet the requirements.
Compatibility study of excipients: In the early stage of formulation development, mixing drugs with different excipients and measuring their melting point changes can preliminarily determine whether there is an interaction between the drug and excipients.
Polycrystalline screening: By measuring the melting points of crystals obtained under different crystallization conditions, possible polymorphic phenomena can be quickly detected, providing clues for further characterization (such as X-ray diffraction, thermal analysis).
Intermediate monitoring: In chemical synthesis processes, melting point determination of key intermediates can quickly determine whether the reaction and product purity meet the standards, thereby optimizing process conditions.
In summary, although drug melting point determination is a traditional technology, its principle is scientific, operation is simple, and results are intuitive. It still plays an irreplaceable role in modern drug analysis and is the first important checkpoint to ensure drug quality.
