Fully automatic transdermal diffusion apparatusBy simulating the human skin environment and combining precise fluid control and detection technology, automated monitoring and analysis of the transdermal absorption process of drug or cosmetic ingredients can be achieved. Its core working principle can be divided into four key steps: skin simulation, diffusion driven, environmental control, and data acquisition. The following is a detailed analysis:
1、 Skin Simulation: Building a Transdermal Barrier Model
Material selection:
Artificial synthetic membranes, such as silicone membranes and polyurethane membranes, simulate the barrier function of the stratum corneum of the skin by adjusting the pore size and thickness, with low cost and good repeatability.
Ex vivo animal skin: commonly used mouse skin, rabbit skin, or pig skin, which is closer to the true skin structure (including stratum corneum and dermis), but requires fresh preparation and individual differences exist.
3D skin model: a skin like tissue constructed based on cell culture technology, including keratinocytes, fibroblasts, etc., suitable for research (such as drug toxicity assessment).
Diffusion pool design:
Vertical (Franz cell): consisting of a supply cell (containing sample) and a receiving cell (containing receiving liquid), with a skin simulation membrane sandwiched in between. The components in the sample diffuse to the receiving solution through the concentration gradient, which is suitable for semi-solid preparations (such as ointment and gel).
Horizontal (Valia Chien cell): Similar in structure, but with a horizontal distribution of sample and receiving liquid, suitable for large-area administration forms such as patches and Babuses, with a larger drug loading capacity.
Special design: such as dual sampling port horizontal transdermal cup (supporting iontophoresis research), nail specific vertical diffusion cell, etc., to meet specific experimental needs.
2、 Diffusion driven: concentration gradient and physical promotion
Passive diffusion:
The core mechanism is driven by concentration gradient: when the concentration of components in the sample is higher than that of the receiving solution, molecules spontaneously diffuse from high concentration areas to low concentration areas, penetrate the skin simulation membrane, and enter the receiving solution.
The diffusion rate is influenced by molecular size, lipid solubility, charge properties, and skin barrier integrity (such as small molecules and lipid soluble components being more easily permeable).
Physics Promotion Technology (optional):
Ion introduction: By driving charged molecules (such as ionic drugs) to move in a directional manner through the action of an electric field, the transdermal rate is significantly improved.
Ultrasonic Introduction: Utilizing ultrasonic waves to generate cavitation effects, temporarily disrupting the structure of the skin's stratum corneum and enhancing permeability.
Microneedle array: By forming microchannels on the surface of the skin with tiny needles, it bypasses the barrier of the stratum corneum and achieves transdermal delivery of macromolecules such as proteins and vaccines.
3、 Environmental control: precise simulation of physiological conditions
Temperature regulation:
Using a dry heating block or circulating water bath system, the diffusion pool temperature is stabilized at 32 ± 0.2 ℃ (simulating the surface temperature of human skin) to avoid temperature fluctuations affecting molecular movement and skin barrier function.
Humidity control:
Maintain the humidity inside the diffusion tank through a humidification module to prevent the skin simulation membrane or detached skin from drying and cracking, ensuring consistency in experimental conditions.
Mixing and Mixing:
The receiving tank is equipped with a magnetic stirring device, which uniformly mixes the receiving liquid at a speed of 50-1200rpm to prevent local concentration from being too high or too low, ensuring that the diffusion process conforms to Fick's law.
4、 Data Collection: Automated Monitoring and Quantitative Analysis
Sampling system:
High precision infusion pump: Extract quantitative samples (with an error of ≤ 0.08ml) from the receiving tank at preset time intervals (such as every hour) to avoid errors introduced by manual sampling.
Multi channel synchronous detection: supports parallel experiments with 6-12 channels, allowing for simultaneous comparison of the effects of different formulations, concentrations, or penetration promotion techniques.
Detection method:
UV Visible spectrophotometry (UV Vis): Suitable for components with UV absorption (such as certain drugs and dyes), the concentration is calculated by absorbance.
High performance liquid chromatography (HPLC): Separate target components from complex samples and quantitatively analyze low concentrations or structurally similar substances.
Mass spectrometry (MS): High sensitivity detection of trace components, suitable for metabolite or stability studies.
Software analysis:
Real time curve drawing: Automatically generate transdermal rate time curve, calculate cumulative permeation rate (Qn) and steady-state transdermal rate (Jss).
Data Management: Supports paperless log queries, historical data backtracking, and local printing, in compliance with GLP (Good Laboratory Practice) requirements.
Permission control: Set four levels of management permissions to prevent unauthorized operations or method modifications and ensure data reliability.
