Detailed explanation of the use of water quality derived heaters
Date: 2025-07-08Read: 1
The water quality derivative heater is a key equipment used in environmental monitoring, water quality analysis, and chemical experiments to promote derivative reactions, especially widely used in trace organic matter analysis (such as high-performance liquid chromatography, gas chromatography pretreatment). The following provides a detailed explanation of equipment principles, operating procedures, parameter control, maintenance points, and safety regulations, aiming to help users standardize operations, improve experimental efficiency, and extend equipment lifespan.
1、 Equipment principle and applicable scenarios
1. Core functions
By precise temperature control heating, the chemical reaction between the derivative agent and the target substances in the water sample (such as amines, phenols, aldehydes, etc.) is accelerated, generating stable products that can be recognized by the detector (such as fluorescent derivatives, UV absorbing products). Common derivative technologies include:
-Fluorescence derivatization method (such as OPA/β - CD reaction with amino acids)
-Acylation reactions (such as EDC and carboxylic acid condensation)
-Esterification reaction (such as BF3 methanol and fatty acid methylation)
2. Typical application scenarios
-Analysis of trace pollutants in environmental water samples (such as polycyclic aromatic hydrocarbons, algal toxins)
-Humus detection in industrial circulating water system
-Determination of residual additives in food and beverages
-Pre treatment of metabolites in biological samples
2、 Operation process and key steps
1. Preparation for preprocessing
-Sample filtration: Use a 0.45 μ m filter membrane to remove particulate matter and avoid clogging the pipeline.
-Derivative preparation: Prepare and use according to standard ratios (e.g. OPA solution needs to be stored away from light), and some reagents require nitrogen blowing to remove oxygen.
-Blank control: Simultaneously prepare a blank sample of pure water and derivatization agent to eliminate background interference.
2. Device initialization
-Installation of reaction tank: Select quartz or polytetrafluoroethylene reaction tubes with appropriate volume to ensure that the sealing ring is not aged or deformed.
-Pipeline connection: The peristaltic pump hose needs to be adjusted to the appropriate tightness (too tight is prone to crushing, too loose is prone to leakage), and the waste liquid tank should be placed below the outlet position.
-Temperature calibration: Use a standard thermometer to verify the displayed temperature, with an error of ≤± 1 ℃ (precision experiments require ≤± 0.5 ℃).
3. Parameter settings
-Temperature control: Set the heating temperature according to the reaction type (common range 40-120 ℃), and the recommended heating rate is ≤ 5 ℃/min to avoid boiling.
-Time adjustment: The reaction time is usually 5-30 minutes, and the timer error should be less than 1 second.
-Flow rate matching: The peristaltic pump speed is linked to the heating time to ensure sufficient mixing of reactants (recommended residence time ≥ 3 times the theoretical reaction time).
4. Derivative reaction execution
-Sample addition sequence: Inject water sample first, then add derivative agent, and finally introduce inert gas (such as argon) to prevent oxidation.
-Mixing method: Use vortex mixing or online mixer to avoid the generation of bubbles.
-Real time monitoring: Observe the color change of the reaction solution (such as fluorescence induced blue) and record the actual reaction time.
3、 Precautions and problem avoidance
1. Temperature deviation control
-Thermal distribution test: The new equipment requires a multi-point thermometer to detect the temperature difference inside the reaction tank, with a requirement of ≤± 2 ℃.
-Preheating treatment: Burn three times before the formal experiment to eliminate the material memory effect.
-Environmental compensation: When the laboratory temperature fluctuates by more than 5 ℃, enable PID temperature control mode.
2. Prevention of cross contamination
-Special pipeline identification: Use independent pipelines for different sample types, and soak in 10% nitric acid for 2 hours when replacing.
-Cleaning procedure: Rinse the system with ultrapure water, methanol, and acetone in sequence after each experiment, and dry for later use.
-Bubble trap setting: Install an exhaust valve at the highest point of the pipeline to prevent air resistance from affecting the flow rate.
3. Derivative efficiency optimization
-PH value adjustment: Alkaline conditions can easily lead to amide hydrolysis, while acidic conditions may inhibit certain derivative reactions, which need to be strictly regulated according to the method requirements.
-Light control: The operation of photosensitive derivatives should be carried out in a brown bottle away from light.
-Vibration frequency: It is recommended to use a constant temperature shaker (200rpm, 45 ° tilt angle) for offline derivation.
4、 Daily maintenance and troubleshooting
1. Periodic maintenance
-Sensor calibration: Calibrate the temperature control probe with a standard platinum resistance thermometer every quarter.
-Seal replacement: It is recommended to check the silicone gasket monthly and replace it immediately if it hardens or deforms.
-Heating block inspection: The surface flatness of the heating block is tested annually (using a feeler gauge method), and any unevenness>0.1mm needs to be polished.
5、 Safety operation standards
1. Protective equipment: Wear nitrile gloves, goggles, and gas masks when handling corrosive reagents.
2. Emergency response: In case of skin contact with derivative agents, rinse immediately with 70% ethanol. If ingested, drink 200ml of milk and seek medical attention.
3. Equipment grounding: Ensure that the grounding resistance of the casing is less than 4 Ω, and use is prohibited during thunderstorms.
4. Waste disposal: Collect heavy metal derivatives according to the hazardous waste code HW49.
