What are the common problems with capillary chromatography columns?
Date: 2025-10-23Read: 0
Capillary chromatography columnThere may be various problems encountered during use, which may affect separation efficiency, analysis accuracy, and even the lifespan of the chromatographic column. Here isCapillary chromatography columnA detailed summary of common problems and their solutions:
1、 Decreased column efficiency
Problem description: The separation ability of the chromatographic column decreases, manifested as broadening of peak shape, decrease in resolution, or reduction in theoretical plate number.
Possible reasons:
Fixed phase loss: High temperature or strong solvent action causes the fixed phase coating to peel off.
Column contamination: Impurities or pre-treatment residues in the sample accumulate inside the column.
Insufficient column aging: The new column is not fully aged, and residual solvents or oligomers affect separation.
Mechanical damage: Column bending, loose pressure cap, or clogged sieve plate.
Solution:
Regular aging: New columns or columns that have not been used for a long time should be aged for several hours at a temperature 20-30 ℃ below the maximum operating temperature.
Avoid high temperatures: Strictly control the column temperature to avoid exceeding the maximum tolerance temperature of the stationary phase.
Sample purification: Use methods such as solid-phase extraction, filtration, or centrifugation to remove particles and impurities from the sample.
Replace sieve plate: If the sieve plate is blocked, a new sieve plate needs to be replaced and the column needs to be reinstalled.
2、 Abnormal peak shape
Problem description: Asymmetric peak shape (trailing or protruding), appearance of splitting peaks or ghost peaks.
Possible reasons:
Excessive injection volume: exceeding the column capacity leads to broadening of peak shape.
Improper injection technique: such as inappropriate split ratio during split injection.
Column contamination: Impurities adsorbed on the surface of the stationary phase affect mass transfer.
Excessive dead volume: There is dead volume in the injector, detector, or connecting tubing.
Solution:
Optimize injection volume: Adjust the injection volume according to the column capacity, usually from nanoupgrading to micro upgrading.
Improve injection technology: adjust the split ratio or use a non split injection mode.
Cleaning the chromatographic column: Rinse the column in reverse with a solvent (such as methanol, n-hexane) to remove contamination.
Check dead volume: Ensure that there are no leaks or dead corners in the injector, detector, and connecting pipelines.
3、 Retention time variation
Problem description: The retention time of the same compound is unstable in different batches or within the same batch.
Possible reasons:
Column temperature fluctuation: Unstable temperature control system leads to changes in retention time.
Unstable carrier gas flow rate: gas path leakage or flow meter error.
Fixed phase property changes: such as fixed phase aging or contamination.
Sample processing differences: Inconsistent pre-processing steps lead to changes in the sample matrix.
Solution:
Stable column temperature: Use a high-precision temperature control system to ensure constant column temperature.
Check the air circuit: Regularly inspect the air circuit connections and replace aging or damaged components.
Regular aging: Avoid excessive aging of fixed phases and use according to the recommended conditions in the manual.
Standardized sample processing: Ensure consistent pre-treatment steps and reduce matrix effects.
4、 Column breakage or leakage
Problem description: Column rupture, loose pressure cap, or leakage at the joint.
Possible reasons:
Improper installation: The column is bent or the pressure cap is not tightened.
Mechanical stress: The column is subjected to external impact during transportation or operation.
Material fatigue: After long-term use, the column material ages.
Solution:
Correct installation: Ensure that the column is installed straight and the pressure cap is tightened without excessive force.
Avoid external forces: Handle with care during transportation and operation to avoid bending or impact of the column.
Regular inspection: Regularly inspect the columns, pressure caps, and joints, and replace aging components in a timely manner.
5、 Ghost Peak and Background Interference
Problem description: Unknown peaks or increased baseline noise appear in the chromatogram.
Possible reasons:
Sample contamination: Impurities or pre-treatment residues are present in the sample.
Injector contamination: Aging of injection needle, liner or spacer.
Carrier gas pollution: The carrier gas contains oxygen, moisture, or hydrocarbon impurities.
Detector contamination: Detector filament aging or ion source contamination.
Solution:
Sample purification: Use methods such as solid-phase extraction, filtration, or centrifugation to remove impurities.
Cleaning the injector: Regularly replace the injection needle, liner, and spacer, and clean the injection port.
Purification of carrier gas: Use high-purity carrier gas and install a carrier gas purification device.
Maintain the detector: Regularly clean the detector and replace aging components.
6、 Improper selection of fixed phase
Problem description: Poor separation effect, unreasonable retention time of target compound or co leakage.
Possible reasons:
Fixed phase polarity mismatch: does not match the properties of the sample (such as non-polar column analysis of polar compounds).
Inappropriate column length or inner diameter: Insufficient separation due to short column length or large inner diameter.
Improper selection of film thickness: Thin film thickness leads to low column capacity, while thick film thickness leads to broadening of peak shape.
Solution:
Choose a stationary phase based on the properties of the sample: non-polar samples use non-polar columns (such as DB-5), and polar samples use polar columns (such as DB-WAX).
Optimize column parameters: Select the appropriate column length (such as 15-60m) and inner diameter (such as 0.18-0.53mm) according to the separation requirements.
Adjust film thickness: Choose the appropriate film thickness (such as 0.1-5 μ m) based on the sample concentration and injection volume.
7、 Shortened column lifespan
Problem description: The chromatographic column fails prematurely and needs to be replaced frequently.
Possible reasons:
Improper operating conditions, such as high temperature, high flow rate, or strong solvents.
Serious sample contamination: Insufficient purification.
Insufficient maintenance: Failure to regularly age or clean the chromatography column.
Solution:
Optimize operating conditions: strictly control column temperature, flow rate, and solvent type.
Strengthen sample purification: Use methods such as solid-phase extraction, filtration, or centrifugation to remove impurities.
Regular maintenance: Aging, cleaning, and storage should be carried out according to the recommended conditions in the manual.
