Therapeutic drug concentration monitoring (TDM) is a common clinical testing item and one of the most widely used applications of LC-MS/MS in clinical testing. The LC-MS/MS detection scheme developed during the development and clinical trial stages of the contract, as well as the accumulated clinical experience, directly guided the implementation of TDM work. At the same time, with the gradual maturity of precision medicine and personalized medication, new requirements have been put forward for the development of TDM.

This article selected five commonly used psychotropic drugs for treating epilepsy (as shown in the table below), established mSPE sample processing methods, and conducted basic methodological validation on them.

Results and Discussion

Using HLB polymer magnetic extractant and PURETON M32 extractor, a batch of 32 samples can be processed by fully automatic solid-phase extraction within 20 minutes. During this period, there is no need for human intervention, and there is no need for positive or negative pressure driven liquid flow operations. One click to start running until processing is complete, and the eluent is directly analyzed by LC-MS/MS. MSPE, as a new automated sample pretreatment method, has good linearity, accurate and precise results, meeting the basic methodological requirements of quantitative analysis.
