Common and special cultivation methods for Arabidopsis thaliana in incubators

Arabidopsis, also known as Arabidopsis thaliana, belongs to the Brassicaceae family and is a biennial herbaceous plant. It has a small plant morphology, with a flat diameter of 5cm and a height of 30cm. The growth area is small and easy to cultivate; The genome is small, with only 5 chromosomes, 157 million base pairs, and 27000 genes; Fast growth rate, short cycle, one generation only takes about 6 weeks; It can self pollinate and also hybridize between different germplasm lines. It has many seeds and can produce thousands of seeds per plant and generation. Therefore, Arabidopsis is one of the model plants in plant science, including genetics and plant development research. In 2000, Arabidopsis became the first plant with a fully sequenced genome.
Simple cultivation in Arabidopsis incubators can be used for research on gene expression, organ development, gene mutations, and more.
1. Firstly, place the Arabidopsis seeds on filter paper, disinfect them with 70% alcohol, and then treat them with * (20% bleach and 0.05% Triton X-100 non-ionic surfactant can also be chosen for 20 minutes of sterilization treatment). Rinse with sterile water 5 times, then dot the seeds onto MS medium, pH5.9, Add 0.9% agar and 2% sucrose.
2. Place the seeds under dark conditions at 4 ℃ for vernalization for about 48-72 hours, and promote germination under long or short day conditions,. Using a PLC controller, edit a simple day night mode and set the lighting time to 16 hours (8 hours in darkness) or 14 hours (10 hours in darkness); Optional Q23+continuous light intensity adjustment device, set the light intensity to 100-150umol/m2. s; Set the indoor temperature to 20-22 ℃ under light conditions and 18-20 ℃ under dark conditions; Optional humidity control system, set indoor relative humidity to 70%.
3. The seedlings take about 7-10 days to emerge, and then they are transplanted into the soil for cultivation. Bottom irrigation is used to water them (utilizing the capillary action of the soil). Before watering, the soil can be dried, and then watered every 7-10 days thereafter.
4. The soil can be prepared in a ratio of 10:1:1 using nutrient soil, vermiculite, and perlite, or mixed directly with compost, vermiculite, and peat. During the growth process, ordinary flower fertilizer can be applied once or not at all.
5. When the plant grows to a certain height, the stem can be cut to produce more inflorescences, which can be used for transformation research or to obtain more seeds.

In order to accelerate the growth rate of Arabidopsis thaliana, after being cultivated in soil, long days can be set. Using a PLC controller, the program can be edited to set 24-hour light intensity, light intensity of 100-250umol/m2. s, and temperature of 22-23 ℃.
Arabidopsis root treatment cultivation can reduce light exposure time by 8-12 hours, increase light intensity by 100-350 umol/m2. s, and maintain temperatures of 22-23 ℃ under light conditions and 18-20 ℃ under dark conditions. The rest of the cultivation conditions are the same as normal cultivation.
