Protein Purification SystemIt is a core equipment used in biological laboratories for separating and purifying proteins. By combining chromatography technology, it achieves the goal of obtaining high-purity target proteins from complex samples such as cell lysate and fermentation broth. It is widely used in fields such as biopharmaceuticals, enzyme engineering, and molecular biology research.
Protein Purification SystemComposition:
High pressure pump: provides accurate flow rate control to ensure stable flow of samples in the chromatography column.
Column chromatography: filled with a specific medium to achieve separation based on the physical and chemical properties of proteins.
Detector: Integrated continuous wavelength ultraviolet detector and conductivity detector, supporting multi wavelength synchronous monitoring, real-time tracking of protein elution process.
Collector: Automatically collect the components corresponding to elution peaks, divided into two modes: branch collection and peak collection. Parameters such as collection volume and peak threshold can be set to avoid loss of target proteins.
Control software: Built in chromatography method database, supports simple programming, expert assisted software, and automatic data processing to generate experimental reports.
Working Principle:
Based on chromatography technology, separation of biomolecules is achieved by utilizing the adsorption differences of different fillers. For example, gel filtration chromatography separates proteins according to molecular size, ion exchange chromatography separates proteins according to charge properties, and affinity chromatography uses specific binding of target proteins with specific ligands for purification.
Daily operation standards
Before the experiment, rinse the entire flow path with deionized water and equilibrate the chromatography column until the baseline is stable before injection; The mobile phase needs to be filtered through a 0.22 μ m membrane and degassed to avoid bubbles affecting detection and column efficiency.
During gradient elution, avoid sudden changes in the pH value or salt concentration of the mobile phase to prevent denaturation of the target protein or damage to the chromatography medium.
Immediately rinse the chromatography column with a suitable eluent after the experiment, equilibrate with deionized water or preservation solution, seal and store in refrigeration.
