In biological research and drug discovery, there is an increasing need to expand the diversity and complexity of cell analysis. Stem cell-derived cells and tissues are increasingly becoming an alternative to traditional in vitro and in vivo experiments in drug development and toxicological safety assessment. In this study, we used human iPSC derived myocardial and neuronal cell models to develop functional and morphological detection results, and tested the effects of different compounds in a multi parameter analysis format. We use the automated cell imaging and analysis ImageXpress Pico personalized high content imaging analysis system to simultaneously determine calcium oscillation frequency, cell viability, cytoskeleton integrity, cell apoptosis, and mitochondrial function. Measure the effect of calcium oscillation on the beating frequency of myocardial cells. Neurosphere automatic imaging is used to evaluate the number of live cells and to monitor calcium oscillations using time-lapse imaging. The multi type composite evaluation of different detection results provides a deeper understanding of the mechanisms of action of various compounds. The experiment used a set of cardiac active drugs, neurotransmitters, and selected neurotoxic or cardiotoxic compounds for characterization and analysis. Overall, our results demonstrate how various analyses can be utilized to quantitatively screen chemical effects in iPSC cardiomyocytes and neuronal models, and achieve rapid and cost-effective multidimensional biological mapping.
