Ultrasonic extraction equipment is an efficient tool in modern natural product extraction, drug development, environmental analysis, and food testing. It is widely used for the extraction of plant active ingredients (such as flavonoids, alkaloids, polysaccharides), active substances in traditional Chinese medicine, oils, and pollutants. The core principle of ultrasonic extraction equipment is to utilize the "cavitation effect" generated by ultrasonic waves in liquids - tiny bubbles rapidly generate, grow, and violently collapse, instantly releasing high temperature and high pressure energy, damaging cell walls or tissue structures, accelerating solvent penetration and target component dissolution.


Step 1: Equipment inspection and environmental preparation
Confirm that the ultrasonic generator, transducer, and extraction tank are securely connected and the power supply voltage matches. The equipment should be placed on a stable, ventilated, and dry experimental platform, away from moisture and corrosive gases. Check if the cooling system (such as circulating water cooling) is functioning properly to avoid overheating of the transducer.
Step 2: Sample pretreatment and solvent selection
Crush the materials to be extracted (such as medicinal herbs, fruits and vegetables, soil) to a suitable particle size (usually 40-80 mesh) and increase the contact area. Select suitable solvents (such as ethanol, methanol, water, acetone) based on the polarity of the target components, and ensure the purity of the solvents. The amount of solvent used is generally 10-15 times that of the material.
Step 3: Sample installation and sealing operation
Mix the material with the solvent and pour it into the extraction tank. The liquid level should be lower than 2/3 of the tank volume, leaving space for cavitation. No no-load operation is allowed to prevent damage to the transducer. Tighten the can lid to ensure a good seal and prevent solvent evaporation or leakage.
Step 4: Parameter Setting and Program Startup
Set key parameters through the control panel:
Ultrasonic power: Adjust according to material hardness and solvent properties (usually 30% -80% rated power) to avoid excessive degradation of components.
Frequency: Typically 20-40kHz, some devices support multi frequency or variable frequency to improve extraction uniformity.
Time: usually 20-60 minutes, can be divided into sections to avoid overheating caused by prolonged continuous work.
Temperature: If equipped with temperature control function, set a suitable extraction temperature (such as 40-60 ℃) to prevent the deactivation of thermosensitive components.
Step 5: Operation monitoring and process observation
After starting the device, observe whether the liquid produces obvious cavitation phenomena (such as misty bubbles, hissing sounds). Monitor temperature changes inside the tank and activate the cooling system if necessary. Avoid frequent opening of the can lid to prevent sudden pressure changes and solvent loss.
Step 6: Extraction completion and sample processing
After the program is completed, turn off the power and wait for the system to cool down before opening the can lid. Filter or centrifuge the extract to separate the residue. The extract can be further concentrated, purified, or directly analyzed.
