The working principle of Hamilton ion exclusion chromatography column:
1. Donnan exclusion effect: Ion exclusion chromatography is based on the Donnan membrane equilibrium principle, and the stationary phase usually uses high-capacity sulfonated cation exchange resin with a fixed charge on its surface. When the sample solution flows in, the strongly ionized component will quickly pass through the chromatographic column due to repulsion of the same charge, with almost no retention; Non ionic or weakly ionized substances, on the other hand, are not affected by this repulsive force and can enter the resin for distribution.
2. Multi mechanism synergistic separation: In addition to Donnan repulsion, it also includes adsorption and spatial exclusion. Weak ionic substances are retained through non-ionic interactions such as van der Waals forces and hydrophobic interactions, which enable effective separation of compounds with different properties.
3. Selection of mobile phase: Dilute hydrochloric acid or solutions containing organic acids are usually used as the mobile phase to further promote the rapid elution of ionized substances while maintaining the retention behavior of non-ionic components. The detection system is often equipped with conductivity detectors and UV visible light detectors to meet the identification needs of different properties of the analyte.
