Chromatography column is the main instrument for chromatographic analysis, and the correct installation of chromatography column can ensure its optimal performance and prolong its service life. Please refer to the following steps for correct installation:Step 1. Check the gas filter, carrier gas, injection pad, and liner to ensure smooth and effective use of auxiliary gas and detector gas. If dirty samples or highly active compounds have been prepared before, the liner of the injection port needs to be cleaned or replaced.
Step 2. Install the nut and gasket onto the chromatography column, and carefully cut both ends of the column flat
Step 3. Connect the chromatographic column to the injection port. The depth of insertion of the chromatographic column into the injection port depends on the GC instrument used. Correct and appropriate insertion can ensure the reproducibility of test results to the greatest extent possible. Generally speaking, the inlet of the chromatographic column should be kept in the middle and lower part of the injection port. When the injection needle passes through the spacer * and is inserted into the injection port, if the needle tip is 1-2cm away from the inlet of the chromatographic column, this is the ideal state. (For specific insertion degree and method, refer to the random manual of the GC used) Avoid forcefully bending and squeezing the capillary column, and be careful not to let sharp edged objects such as markers come into contact with and rub against the capillary column to prevent the column body from breaking or being damaged. After correctly inserting the chromatographic column into the injection port, tighten the connecting nut by hand. After tightening (which cannot be done by hand), use a wrench to tighten an additional 1/4-1/2 turn to ensure the sealing degree of the installation. Due to loose installation, not only can the device leak, but it may also cause damage to the chromatography column.
Step 4. Connect the carrier gas. After the chromatographic column is connected to the injection port, introduce the carrier gas and adjust the pre column pressure to obtain the appropriate carrier gas flow rate
