What are the procedures to pay attention to when installing liquid chromatography columns
Date: 2025-08-26Read: 0
The following is a detailed description of the installation method for liquid chromatography columns:
Preliminary preparation
-Check instrument status: Turn on the liquid chromatograph, confirm that there are no alarm prompts in the system, and that all components are operating normally. Pay special attention to the pressure stability of the pump and the baseline stability of the detector, which are prerequisites for subsequent installation and use.
-Check the appearance of the chromatography column: New liquid chromatography columns usually come with a factory inspection report and certificate of conformity. Check if there are any damages, cracks, or other issues on the column body of the chromatography column, especially at the joint parts at both ends of the column. If any problems are found, they should be replaced in a timely manner. At the same time, record the model and serial number of the chromatographic column for future reference and maintenance.
Installation steps
1. Emptying the residual mobile phase in the system: Discharge the existing mobile phase inside the instrument through multiple valves or other means to avoid mixing with the storage solvent in the new chromatographic column and producing precipitates or interfering substances. The pump can be started to run at a lower flow rate for a period of time to achieve this operation.
2. Connect the chromatographic column: Determine the direction of the mobile phase inlet and outlet of the chromatographic column (usually the manufacturer will have clear markings on the chromatographic column). Carefully connect one end of the chromatographic column to the tubing behind the injector and the other end to the tubing in front of the detector. When connecting, it is necessary to ensure that the interface is tightly fitted. The commonly used method is to first tighten the connecting nut by hand, and then use a wrench to reinforce it appropriately, but pay attention to moderate force to avoid damaging the chromatographic column joint.
3. Initial flushing: After installation, select a suitable initial flow phase for flushing the chromatographic column. For reverse phase chromatography columns, a high proportion of water organic solvent mixed solution is often used as the initial mobile phase. Set a lower flow rate to allow the mobile phase to slowly flow through the chromatography column, with the aim of replacing any residual storage solvent inside the column and wetting the surface of the stationary phase packing to eliminate bubbles.
4. Equilibrium chromatography column: After initial flushing, switch to the target mobile phase for equilibrium operation before analysis. Stabilize the chromatographic column under the target mobile phase conditions and observe pressure changes and baseline conditions. When the pressure is stable and the baseline tends to flatten, it indicates that the chromatographic column is basically in equilibrium and sample analysis can be performed.
Precautions
-Avoid introducing bubbles: During the entire installation process, special attention should be paid to preventing air from entering the chromatography column. Because bubbles can occupy the space inside the column, affecting the separation efficiency of solutes, and may also cause local high pressure damage to the chromatographic column. So during the process of connecting pipelines and flushing, it is necessary to always pay attention to whether there are bubbles generated, and if so, they should be eliminated in a timely manner.
-Control flow rate and pressure: When flushing and balancing at the beginning, it is not advisable to use excessively high flow rates to avoid loosening or shifting of the chromatographic column bed due to sudden high pressure shocks. As the chromatographic column gradually adapts to the mobile phase environment, adjust the flow rate to the appropriate range according to the actual situation.
By following the installation steps and precautions outlined above, it is possible to ensure the correct installation and use of the liquid chromatography column, laying the foundation for obtaining accurate and reliable analysis results.
