Uneven suction of multi-channel pipette: a hidden obstacle to precision experiments
Date: 2025-11-22Read: 1
In precision experiments such as molecular biology and cell culture,Multi channel pipettes have become standard equipment in laboratories due to their ability to process multiple samples simultaneously. However, the common problem of uneven liquid absorption is like an "invisible danger", which can lead to an increase in the dispersion of experimental data in mild cases and directly affect experimental conclusions in severe cases. This article will systematically explore the key issue that troubles researchers from the analysis of phenomena, tracing the causes to solutions.

1、 Typical manifestations and hazards of uneven liquid absorption
When the multi-channel pipette performs liquid suction operation, if the volume difference of liquid sucked in by each channel exceeds the allowable range (usually ± 3%), it is considered as uneven liquid suction. For example, when configuring a PCR reaction system, if a channel absorbs an additional 1 μ L of primer, it may lead to abnormal amplification efficiency; Uneven liquid absorption during cell plating can directly cause uneven cell density, affecting the results of subsequent differentiation or proliferation experiments. Even more covertly, long-term use of pipettes with uneven liquid absorption may lead to reagent waste, decreased experimental reproducibility, and even question the credibility of the entire research project.
2、 In depth analysis of the four core causes
1. Mechanical structure wear and tear
The aging of piston sealing rings or the attenuation of spring elasticity are the primary causes. Taking a certain brand of 8-channel pipette as an example, after 2000 consecutive uses, micro cracks may appear on the surface of the sealing ring, causing some channels to leak during suction and reducing the suction volume. Spring fatigue will cause inconsistent rebound speed of the piston, further exacerbating the difference in suction.
2. Deviation in operating techniques
Improper control of verticality is the "number one killer" in human factors. Experiments have shown that when the tilt angle of the pipette exceeds 15 °, the error rate of the suction volume can soar to 8%. In addition, if the suction speed is too fast (>2 seconds/channel) or the pressure is not fully released after draining, it can also cause liquid level fluctuations, resulting in inconsistent suction volumes in each channel.
3. Lack of maintenance and upkeep
Unclean pipettes are prone to channel blockage due to residual reagent crystallization or biological contamination. For example, residual DNA enzymes may corrode metal parts, while protein precipitation can alter piston movement resistance, ultimately resulting in uneven liquid absorption.
4. Design defects in adaptability
Some universal pipettes have compatibility issues when matching different specifications of suction tips. If the taper of the suction head does not match the nozzle, it may result in poor sealing, especially when dealing with high viscosity liquids such as glycerin, where uneven suction is more pronounced.
3、 Systematic solution: from prevention to correction
1. Standardized operating procedures
Develop the "Three Verticals Principle": when suctioning, the gun body should be vertical, the suction head should be vertically immersed in the liquid surface, and when draining, the gun head should be vertically aligned with the container wall. It is recommended to adopt a "slow suction and fast discharge" rhythm, which means that during the suction stage, it takes 2-3 seconds to slowly extract and release pressure instantly during the discharge.
2. Hierarchical maintenance system
Establish a three-level maintenance system of daily inspection, weekly cleaning, and monthly calibration: check the integrity of the sealing ring before daily use; Wipe the gun body with 75% ethanol every week and lubricate the piston; Send professional institutions for calibration every month, with a focus on testing the consistency of liquid absorption in each channel.
3. Application of intelligent assistive technology
Some models are equipped with pressure sensors and LCD screens, which can monitor the suction pressure of each channel in real time and automatically compensate for errors.
4. Optimization of consumables matching
Choose a certified pipette tip that matches the brand of the pipette and avoid using third-party compatible products. For high-precision experiments, pre sterilized low adsorption tips can be used to reduce the impact of liquid residue on volume.
The suction level accuracy of multi-channel pipettes is not only a reflection of tool performance, but also the cornerstone of experimental scientificity. Through the synergy of structural optimization, operational standards, and intelligent technology, we can control the unevenness rate of liquid absorption within 1%, providing reliable support for cutting-edge research such as precision medicine and gene editing.
