Automatic peptide synthesizerAs the core equipment for peptide drug development, its long-term stability relies on scientific maintenance plans and regular calibration. Reasonable maintenance not only extends the lifespan of the instrument, but also ensures the purity and sequence accuracy of the synthesized product.
1、 Daily and periodic maintenance
Daily: After running, execute the standard cleaning procedure (DMF → methanol → nitrogen drying); Check if the waste liquid tank is overflowing; Visually confirm that there is no leakage.
Weekly: Replace the sealing gasket of the reagent bottle and the injection needle; Clean the reaction column bracket; Check the nitrogen pressure (recommended 0.4-0.6 MPa).
Monthly: Ultrasonic cleaning of reaction column bottom sieve plate; Check the wear of the peristaltic pump roller; Test the response of all solenoid valve actions.
Every 6 months: replace all flow PEEK pipes (especially at bends); Calibrate temperature sensor (within ± 1 ℃ of heating module); Verify the deprotection efficiency (tested with ninhydrin).
2、 Annual calibration core project
Liquid distribution accuracy calibration: Use a high-precision balance to weigh the solvent mass output from each channel, with an error of ≤± 2%;
Temperature control system calibration: Place the standard PT100 probe at the reaction column position and compare the deviation between the set value and the measured value;
Nitrogen flow calibration: Use an electronic flow meter to detect the blowing airflow and ensure stability at 50-100 mL/min;
Software and hardware synchronization testing: Simulate the complete synthesis process, verify valve switching, pump start stop, and program instruction consistency.
3、 Precautions
Calibration should be performed by manufacturer engineers or certified technicians, and a calibration report should be issued;
All maintenance operations must be carried out under power-off and pressure relief conditions;
It is recommended to establish a "Equipment Use and Maintenance Log" to record each malfunction, consumable replacement, and calibration results for easy traceability and auditing (especially in GMP environments).
