In the process of artificial insemination of pigs, the pig sperm microscope can select high-quality semen for artificial insemination through strict detection of semen quality, thereby improving the conception rate and litter size of sows. For example, in large pig farms, technicians use this microscope to test multiple pig semen samples every day, and only semen that meets quality standards will be used for artificial insemination.
Steps for using a pig sperm microscope:
1. Preparation work:
-Place the microscope on a stable surface, ensuring that it does not shake. Check whether the various components of the microscope are intact, such as whether the lens is clean, whether the objective lens and eyepiece are correctly installed, etc.
-Adjust the light source of the microscope to a moderate brightness, which allows for clear observation of the sample without damaging the eyes or affecting the activity of the sample due to excessive light.
2. Sample processing:
-Side hold the pig essence bag with the detection point facing downwards, ensuring that no bubbles remain at the detection point.
-Squeeze the semen from the tail detection area of the pig sperm bag back into the bag.
-Flip and mix 5 times, gently press the pig sperm bag up 5 times to rinse the remaining semen in the detection area, and squeeze it down to half fill the detection area, then clamp the connection between the detection areas.
-Preheat the detection area in a heating plate or constant temperature water bath at 43 ℃ for 3-5 minutes.
3. Observation and counting:
-Place the processed sample on the microscope stage and secure the position with a clamp to prevent sample movement.
-First, use a low magnification mirror to locate the sample area, and then gradually switch to a high magnification mirror for observation.
-Observing the morphology, vitality, and other conditions of sperm, a counting board can be used to count the number of sperm.
Precautions for using pig sperm microscope:
1. In terms of microscope operation:
-When adjusting the focal length and magnification, operate slowly and gently to avoid excessive force that may damage the lens or cause sample displacement.
-After use, turn off the light source of the microscope in a timely manner to extend the service life of the light bulb. Regularly clean and maintain the microscope to keep the lens and body clean.
2. In terms of sample processing:
-When processing samples, it is necessary to strictly follow the operating steps to avoid introducing impurities or damaging the integrity of the sample.
-Pay attention to temperature control of the sample, and ensure accurate preheating temperature and time to ensure that the sample is in a suitable observation state.
3. In terms of safety protection:
-If using equipment such as electron microscopes, attention should be paid to preventing electric shock accidents and ensuring good grounding of the equipment.
-During the processing of samples and the use of microscopes, protective equipment such as gloves should be worn to avoid contact with harmful substances or contamination by the samples.
