(1) Principles of Optical Imaging
1. Light source system
-Pig sperm microscopes are usually equipped with suitable lighting devices, such as halogen lamps or LED lamps. These light sources can provide bright and stable light to illuminate pig semen samples placed on glass slides. Light is focused and adjusted through optical components such as a condenser to ensure uniform illumination of the sample, providing a foundation for clear observation.
2. Objective lens and eyepiece system
-The objective lens is a group of lenses located near the sample, which have different magnifications. After the light passes through the pig semen sample, the objective lens preliminarily magnifies the tiny structures in the sample, forming an inverted real image. This real image is then further magnified by an eyepiece, which acts like a magnifying glass to enlarge the image formed by the objective lens. The eyepiece also adjusts the clarity and field of view of the image, allowing the observer to see a clear and magnified pig sperm form.
-For example, under a low-power objective lens, the approximate distribution and overall morphology of sperm in pig semen can be observed, such as the rough outline of the sperm's head and tail; When switching to a high magnification objective, the details of the sperm head, such as the shape of the acrosome and the outline of the nucleus, can be seen more clearly.
(2) Sample preparation and observation
1. Sample preparation
-For pig semen samples, it is first necessary to dilute them appropriately. This is because pig semen itself is relatively viscous, and direct observation may affect vision and sperm activity. Specific physiological saline or other buffer solutions are usually used to dilute semen, allowing the sperm to be evenly distributed on a glass slide.
-Then, drop the diluted semen onto a glass slide and cover it with a cover slip. The function of a cover glass is to prevent the sample from drying out, avoid contamination from external impurities, and ensure that the sample is at a suitable thickness for light transmission and microscopic observation.
2. Observation process
-Place the prepared glass slide on the stage of the microscope, and gradually make the sample clearer by adjusting the coarse and fine focus helices. During the observation process, the observation area and contrast can be selected and adjusted by moving the glass slide or adjusting the field of view aperture of the microscope, in order to better observe the morphology, vitality, and other characteristics of pig semen.
